Posters., 2024 08 Assessment of the biological impact of Taraxacum officinale on alpine ecosystems in Japan and extermination methods ―Alien Dandelion Research Team―, NANAMI Satoshi; TOKUOKA Jumpei; MATSUYAMA Shuhei; ITOH Akira, Annual Report of Pro Natura Foundation Japan, 33, 2024 05 30 , 10.32215/pronatura.33.0_51, Researches on Taraxacum officinale, an invasive plant in Japan, have been conducted in urban areas and their suburbs where human activities are high. However, it has been recently reported that T. officinale has invaded alpine areas in Japan, where the influence of humans was minimal. In this study, we investigated the invasion of T. officinale and hybrids between the alien and the Japanese native one in three alpine regions: Mt. Ibuki (1,377 m), Mt. Hakusan (2,702 m) and the Ushirotateyama Mountain Range (2,932 m). In the three alpine regions, the alien and hybrid dandelions were found growing in places under anthropogenic disturbance, such as parking lots and gondola stops. Approximately half of the individuals judged to the alien dandelions based on their morphology were hybrid dandelions, and the proportion of hybrids tended to rise with increasing the altitude. In Mt. Ibuki and Mt. Hakusan, the clonal diversity of the invasive dandelions was low, and a limited number of clones tended to increase the population. On the other hand, the diversity of clones in the Ushirotateyama Mountain Range was high, suggesting that clones of invasive dandelions with various genotypes have established. 日本学術振興会, 科学研究費助成事業, アポミクシス外来植物の遺伝的変異と侵入時期から定量する無性生殖種の遺伝的進化速度, 20230401, 20260331, 松山 周平; 伊東 明; 名波 哲 Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Elucidation of the mechanism of soil nitrogen dynamics in consideration of diversity of mycorrhizal type of tree-understory vegetation in forests, 20220401, 20250331, 福澤 加里部; 谷口 武士; 舘野 隆之輔; 智和 正明; 菱 拓雄; 松山 周平; 木庭 啓介; 野村 睦, 北海道北部に位置する北海道大学中川・天塩研究林内の天然性冷温帯林、北海道東部に位置する九州大学北海道演習林(足寄)および京都大学北海道研究林標茶区にてミズナラ対象木の周囲にプロットを設定し、ササ除去区と隣接する対照区との間で比較した。このうち中川・天塩での解析を中心に進めた。中川サイトではクマイザサ、天塩の天然林サイトではチシマザサ、足寄サイトではミヤコザサが林床に密生していた。また天塩では皆伐跡地のクマイザサ群落においても調査をおこなった。2022年7月~8月に表層10㎝土壌を採取し、アンモニウム態・硝酸態窒素量 (NH4-N, NO3-N) を測定した。また室内培養 (30℃・28日) 後に同様に測定し、初期値との差から正味のNH4-N・NO3-N生成ポテンシャルとその合計の窒素無機化ポテンシャルを定量した。北海道北部においては、全体的にNH4-Nが土壌無機窒素の優占形態であった。土壌中のNH4-N, NO3-N量に処理区間で有意な差はなかった。ササ除去処理は正味のNO3-N生成ポテンシャルに負の影響を及ぼし、特にササ除去からの時間経過が長い天塩サイトで顕著であった。ササ消失は中長期的には無機窒素の特にNO3-N生成に抑制的に作用することが示唆された。また、単位面積あたりのササの地上部バイオマス、稈高、稈密度、林床の光環境(rPPFD)を調べた。ササバイオマスは足寄(ミヤコザサ)よりも天塩・中川(クマイザサ・チシマザサ)で有意に大きかった。ササバイオマスは稈高、稈密度と正の相関があったが、稈高と稈密度の効果の大きさは足寄と天塩・中川とは異なっており、優占するササの種類の違いがバイオマスの差に影響していることが示唆された。 Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research Grant-in-Aid for Scientific Research (B), Clarification of mechanisms of soil nitrogen dyanamics considering mycorrizal type of trees-understory vegetation system, 20220401, 20250331, 福澤 加里部; 谷口 武士; 舘野 隆之輔; 智和 正明; 菱 拓雄; 松山 周平; 木庭 啓介; 野村 睦 Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research Grant-in-Aid for Scientific Research (B), Invasion process of apomictic plants: a case study of hybrid dandelions in Japan, 20180401, 20220331, Itoh Akira, The objective of this study is to clarify the process of establishment and distribution expansion of hybrid dandelions between asexually reproducing alien species, Taraxacum officinale, and sexually reproducing native species, e.g., T. japonicum and T. platycarpum. We 1) examined the 10-year changes in the ratio of hybrids to T. officinale in western Japan, 2) estimated the origin(s) and migration routes of hybrids, 3) investigated the distribution status of hybrid dandelions in Hokkaido, where few hybrids had been recorded, and 4) verified if various hybrid clones were adapted to different environments. As a result, it was estimated that hybrid dandelions have been formed repeatedly in various regions in Japan, creating various clones that adapted to new environments, and that these hybrids have expanded their distribution through asexual reproduction. In one brown bear habitat area, mountain grape (Vitis coignetiae) seeds were established more than 10 km away, suggesting that there is long-distance seed dispersal in brown bear habitat areas. Because we could not successfully estimate seed dispersal distances in areas without brown bear habitat, we did not know whether the presence or absence of brown bears affects the distance of seed dispersal in V. coignetiae. However, because there were no distinct differences in the genetic diversity of mountain grape between areas inhabited by brown bears and areas not inhabited by brown bears, we inferred that, at least on long time scales, differences in animal seed disperser communities do not affect the rate of plant dispersal. Individuals at the same latitude were genetically similar, and individuals at different latitudes showed different germination/flowering characteristics, suggesting that adaptive evolution is more important than trait plasticity. When individuals of the same clone were grown at high temperature, epigenetic mutations increased, and the mutations were transmitted to the next generation, indicating a possible contribution of epigenetic mutations. No evidence was obtained for hybridization to genetic population diversity as no differences were found in the genetic diversity of T. officinale and hybrid populations. Using a detailed molecular phylogeny estimated for 699 tree species in a Bornean rain forest, we found that maximum size and demographic traits, such as mortality and growth rates, were more similar among phylogenetically more closely related species, while habitat niche was various even among them. This suggests that niche habitat is an evolutionary labile trait and that divergence in habitat niche may enhance coexistence of closely related species, which have similar adult size and demographic traits. We also found that the relationship between species and phylogenetic diversity was not straightforward but rather complicated, which suggested that we need to consider both species and phylogenetic diversity simultaneously in biodiversity conservation of tropical rain forest. It was suggested that the effect of the deer exclusion fence for the vegetation growth and biodiversity was smaller (or, slower) in the non-thinning artificial forest of Cryptomeria japonica than in the thinning forest. It seemed to be the highest (fastest) in the grassland. It was suggested that the overgrazing indirectly affected to the material cycling in the soil by the physiological feature of plant (nitrate reducing enzyme activity), while it directly affected to the plant biodiversity. It indicated that the linkage among the dietary habit of the deer, vegetation and the soil material cycling was important for understanding and evaluating effects of the overgrazing on the forest ecosystem. We found unique cpDNA types to Taraxacum japonicum or other diploid native dandelions, indicating a possibility to use cpDNA for estimating origin of hybrids. We found only few hybrids that had unique cpDNA to T. japnicum. Proportion of hybrids in apparent exotic dandelions were higher in eastern than western Japan. The cultivation experiment showed that hybrids growing in different vegetation differed in cpDNA and growth features. These results suggest that difference in native dandelions that produced hybrids may influence the frequency, establishment and expansion of hybrid dandelions in Japan. Plant species with different nitrogen utilization capacity showed complex dynamics of inorganic nitrogen in soil. Diversity in the vegetation may not directly affect the soil nitrogen dynamics. However, if the dietary preference of the deer would determine the species composition and biomass, the feeding pressure by Sika deer might indirectly affect the nitrogen dynamics in the soil. Prevailing the unpalatable vegetation, the feeding pressure by Sika deer may suppress the leakage of nitrogenous nutrients from the soil.

Microbe communities in the thinning and un-thinning stands in cool temperate cedar plantation, Matsuyama Shuhei; Fukushima Keitaro; Kawamura Koji; Suzuki Shinya; Sakai Shinjiro; Hirai Takeshi; Yoshioka Takahito, The Japanese Forest Society Congress, 間伐は人工林の適切な維持管理に欠かせない施業であるが、林内の上木・下層植生・土壌のみならず、渓流水質を通じて流域環境にも影響を及ぼすと考えられている。渓流水質の変化は、間伐による土壌の炭素・窒素動態の改変に起因するが、このとき土壌微生物群集にどのような変化が起きているのかについては殆ど知られていない。そこで、土壌細菌群集のDNA分析から、間伐による土壌細菌群集の変化を明らかにすること目的として調査を行った。調査は京都大学芦生研究林の2つのスギ人工林で行った。一方の人工林では昨年6月に間伐を行い、他方は間伐を行わない対照区とした。2つの人工林において5月(間伐前)と11月(間伐後)にそれぞれ5地点から0-10cm間の鉱質土壌を採取した。採取した土壌は篩別後にDNAを抽出し、土壌DNA溶液中の細菌16S rDNAの断片(約1400bp)を増幅した。増幅DNA断片は大腸菌の形質転換により単離し、制限酵素処理による切断パターンから土壌試料ごとの16S rDNA多型を決定した。発表では、間伐前後の16S rDNAの多型パターン・多様性と土壌の炭素・窒素動態をサイト間で比較した結果を合わせて示す。 |

Assessment of the biological impact of Taraxacum officinale on alpine ecosystems in Japan and extermination methods ―Alien Dandelion Research Team―, NANAMI Satoshi; TOKUOKA Jumpei; MATSUYAMA Shuhei; ITOH Akira, Annual Report of Pro Natura Foundation Japan, 33, 2024 05 30 , 10.32215/pronatura.33.0_51, Researches on Taraxacum officinale, an invasive plant in Japan, have been conducted in urban areas and their suburbs where human activities are high. However, it has been recently reported that T. officinale has invaded alpine areas in Japan, where the influence of humans was minimal. In this study, we investigated the invasion of T. officinale and hybrids between the alien and the Japanese native one in three alpine regions: Mt. Ibuki (1,377 m), Mt. Hakusan (2,702 m) and the Ushirotateyama Mountain Range (2,932 m). In the three alpine regions, the alien and hybrid dandelions were found growing in places under anthropogenic disturbance, such as parking lots and gondola stops. Approximately half of the individuals judged to the alien dandelions based on their morphology were hybrid dandelions, and the proportion of hybrids tended to rise with increasing the altitude. In Mt. Ibuki and Mt. Hakusan, the clonal diversity of the invasive dandelions was low, and a limited number of clones tended to increase the population. On the other hand, the diversity of clones in the Ushirotateyama Mountain Range was high, suggesting that clones of invasive dandelions with various genotypes have established. |

Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Elucidation of the mechanism of soil nitrogen dynamics in consideration of diversity of mycorrhizal type of tree-understory vegetation in forests, 20220401, 20250331, 福澤 加里部; 谷口 武士; 舘野 隆之輔; 智和 正明; 菱 拓雄; 松山 周平; 木庭 啓介; 野村 睦, 北海道北部に位置する北海道大学中川・天塩研究林内の天然性冷温帯林、北海道東部に位置する九州大学北海道演習林(足寄)および京都大学北海道研究林標茶区にてミズナラ対象木の周囲にプロットを設定し、ササ除去区と隣接する対照区との間で比較した。このうち中川・天塩での解析を中心に進めた。中川サイトではクマイザサ、天塩の天然林サイトではチシマザサ、足寄サイトではミヤコザサが林床に密生していた。また天塩では皆伐跡地のクマイザサ群落においても調査をおこなった。2022年7月~8月に表層10㎝土壌を採取し、アンモニウム態・硝酸態窒素量 (NH4-N, NO3-N) を測定した。また室内培養 (30℃・28日) 後に同様に測定し、初期値との差から正味のNH4-N・NO3-N生成ポテンシャルとその合計の窒素無機化ポテンシャルを定量した。北海道北部においては、全体的にNH4-Nが土壌無機窒素の優占形態であった。土壌中のNH4-N, NO3-N量に処理区間で有意な差はなかった。ササ除去処理は正味のNO3-N生成ポテンシャルに負の影響を及ぼし、特にササ除去からの時間経過が長い天塩サイトで顕著であった。ササ消失は中長期的には無機窒素の特にNO3-N生成に抑制的に作用することが示唆された。また、単位面積あたりのササの地上部バイオマス、稈高、稈密度、林床の光環境(rPPFD)を調べた。ササバイオマスは足寄(ミヤコザサ)よりも天塩・中川(クマイザサ・チシマザサ)で有意に大きかった。ササバイオマスは稈高、稈密度と正の相関があったが、稈高と稈密度の効果の大きさは足寄と天塩・中川とは異なっており、優占するササの種類の違いがバイオマスの差に影響していることが示唆された。 |

We 1) examined the 10-year changes in the ratio of hybrids to T. officinale in western Japan, 2) estimated the origin(s) and migration routes of hybrids, 3) investigated the distribution status of hybrid dandelions in Hokkaido, where few hybrids had been recorded, and 4) verified if various hybrid clones were adapted to different environments. As a result, it was estimated that hybrid dandelions have been formed repeatedly in various regions in Japan, creating various clones that adapted to new environments, and that these hybrids have expanded their distribution through asexual reproduction. |

In one brown bear habitat area, mountain grape (Vitis coignetiae) seeds were established more than 10 km away, suggesting that there is long-distance seed dispersal in brown bear habitat areas. Because we could not successfully estimate seed dispersal distances in areas without brown bear habitat, we did not know whether the presence or absence of brown bears affects the distance of seed dispersal in V. coignetiae. However, because there were no distinct differences in the genetic diversity of mountain grape between areas inhabited by brown bears and areas not inhabited by brown bears, we inferred that, at least on long time scales, differences in animal seed disperser communities do not affect the rate of plant dispersal. |

Individuals at the same latitude were genetically similar, and individuals at different latitudes showed different germination/flowering characteristics, suggesting that adaptive evolution is more important than trait plasticity. When individuals of the same clone were grown at high temperature, epigenetic mutations increased, and the mutations were transmitted to the next generation, indicating a possible contribution of epigenetic mutations. No evidence was obtained for hybridization to genetic population diversity as no differences were found in the genetic diversity of T. officinale and hybrid populations. |

Using a detailed molecular phylogeny estimated for 699 tree species in a Bornean rain forest, we found that maximum size and demographic traits, such as mortality and growth rates, were more similar among phylogenetically more closely related species, while habitat niche was various even among them. This suggests that niche habitat is an evolutionary labile trait and that divergence in habitat niche may enhance coexistence of closely related species, which have similar adult size and demographic traits. We also found that the relationship between species and phylogenetic diversity was not straightforward but rather complicated, which suggested that we need to consider both species and phylogenetic diversity simultaneously in biodiversity conservation of tropical rain forest. |

It was suggested that the effect of the deer exclusion fence for the vegetation growth and biodiversity was smaller (or, slower) in the non-thinning artificial forest of Cryptomeria japonica than in the thinning forest. It seemed to be the highest (fastest) in the grassland. It was suggested that the overgrazing indirectly affected to the material cycling in the soil by the physiological feature of plant (nitrate reducing enzyme activity), while it directly affected to the plant biodiversity. It indicated that the linkage among the dietary habit of the deer, vegetation and the soil material cycling was important for understanding and evaluating effects of the overgrazing on the forest ecosystem. |

We found unique cpDNA types to Taraxacum japonicum or other diploid native dandelions, indicating a possibility to use cpDNA for estimating origin of hybrids. We found only few hybrids that had unique cpDNA to T. japnicum. Proportion of hybrids in apparent exotic dandelions were higher in eastern than western Japan. The cultivation experiment showed that hybrids growing in different vegetation differed in cpDNA and growth features. These results suggest that difference in native dandelions that produced hybrids may influence the frequency, establishment and expansion of hybrid dandelions in Japan. |

Plant species with different nitrogen utilization capacity showed complex dynamics of inorganic nitrogen in soil. Diversity in the vegetation may not directly affect the soil nitrogen dynamics. However, if the dietary preference of the deer would determine the species composition and biomass, the feeding pressure by Sika deer might indirectly affect the nitrogen dynamics in the soil. Prevailing the unpalatable vegetation, the feeding pressure by Sika deer may suppress the leakage of nitrogenous nutrients from the soil. |
