On the other hand, in normal tendon, a few fibroblastic cells and vascular components lied in the endotenon barely expressed its mRNA, but other cellular components in the tendon bundle were not positively hybridized. Tendon tissues for tissue culture were isolated from Achilles tendons of rabbits. The tendon segments were placed one segment per well and incubated in growth medium consisting of Dullbecco's modified Eagle's medium supplemented with 5% fetal bovine serum at 37 degrees C in a humidified atmosphere with 5% CO2 for various periods. The alignment of collagen fibrils was preserved for 48 h, but tendon structure has disintegrated at 96 h. Alcian blue staining and gelatine zymography revealed that proteoglycan markedly diminished and that matrix metalloproteinase (MMPs) activity was upregulated sharply at 72 and 96 h. The ratio of collagen fibrils with large diameter had increased and the mean diameter and mass average diameter value had reached maximum at 48 h. The values then decreased and mean diameters at 72 and 96 h were significantly different from that at 48 h. At 96 h, the ratio of collagen fibrils with small diameters had increased and collagen fibrils with large diameters had disappeared. These findings indicate that structural alteration is possible to be induced by disintegration of collagen fibrils and disappearance of glycosaminoglycans from extracellular matrix (ECM), subsequent of upregulation of MMPs activity. Ingestion of collagen peptide or lactalbumin induced a significant increase in collagen fibril diameter and a decrease in fibril density except for a high dose of lactalbumin compared with the water control. A histogram pattern of fibril diameter in a high dose of collagen peptide showed a peak at 160-180 nm, which was not observed in other groups. However the percentage of diameters over 200 nm was the lowest in this group but highest in the low-dose group of collagen peptide. The mean fibril diameter and mass average diameter of a high dose of collagen peptide were significantly smaller than those in a low dose. The amount of dermatan sulphate increased in the high-dose groups, while the amount of hyaluronic acid decreased in rabbits fed with collagen peptide or lactalbumin at either dose. These results suggest that the ingestion of collagen peptide affects the size of collagen fibrils and composition of glycosaminoglycans in the Achilles tendon and thus may improve the mechanical properties of the Achilles tendon. Samples were obtained from normal and inflamed SDFTs of horses. To detect apoptosis and to identify apoptotic cells, the samples were subjected to immunohistochemical labelling and Western blot analysis. Although a large number of cells in degenerate areas showed positive reactions with caspase-3 and single stranded DNA antibodies, cells in normal tendon samples showed very weak reactions. Excessive apoptosis was confirmed by the results of Western blot analysis, which showed a significant increase in activated caspase-3 protein in the inflamed SDFTs, suggesting that apoptosis occurred in the tendinocytes via a caspase-3-dependent pathways This is the first report of excessive apoptosis in inflamed SDFT of the horse. In this study, we focused on four signaling factors, TNFα, TNF-R1, TNFR-associated factor2 (TRAF2) and caspase-3, in order to elucidate the signaling events in tendinocytes. Samples were obtained from normal, inflamed and scar-formed equine superficial digital flexor tendons. To detect these signaling factors, samples were subjected to immunohistochemistry and Western blot analysis, and some samples were also subjected to reverse transcription-polymerase chain reaction (RT-PCR), PCR-Southern blot analysis and in situ hybridization to detect the expression of TNFα mRNA. Distribution of the four factors differed depending on the tendon condition, normal, inflamed or scar-formed. In the normal tendon, large amounts of TRAF2 were found in tendinocytes, but the amounts of TNF-R1 were small. TNFα mRNA was expressed most highly in the inflamed tendon. TNF-R1, which was only faintly detected in the normal tendon, was detected at a high level in the inflamed tendon, and the amounts of TRAF2 and caspase-3 also increased. Activated caspase-3 was only detected in the inflamed tendon. TNFα mRNA was also expressed in the scar-formed tendon, though it showed weak signals, and the expression levels of TNF-R1, TRAF2 and caspase-3 proteins were very low. Two distinct intracellular signaling pathways of TNFα, which lead to cell survival and apoptosis, might be present in tendinocytes mediated through TNF-R1. These results, which reflect the dynamism of TNFα, provide important clues for means to prevent tendinopathy.

In this study, the localizations of 4 cytokines (IL-1alpha, IL-1beta, TNFalpha and IFNgamma) in tendinocytes of the equine superficial digital flexor tendon (SDFT) were analyzed by the use of an immunohistochemical method. In inflamed tendons positive staining for all 4 cytokines antibodies were detected in endotedinieum cells and vascular epithelial cells. In contrast, negative or trace immunoreactions were obtained in many tendinocytes in the normal tendon. Type-A Sertoli cells at stage V of the spermiogenetic cycle and type-B Sertoli cells at stage Vu of the spermiogenetic cycle were used for analysis. Morphologically, the Sertoli cell processes were classified into sheet-like and slender cord-like processes. The sheet-like process originated solely from the Sertoli cell column while the slender cord-like process projected either from the Sertoli cell column or the sheet-like process. Periodic acid (PA)-thiocarbohydrazide (TCH)-silver protein (SP)-physical development (PD)-positive granules were found diffusively both in the sheet-like and slender cord-like processes near the round spermatid, whereas they had accumulated near the head of the elongated spermatid. The morphological variation and glucoconjugate histochemical reaction of the Sertoli cell processes reflect nourishment, movement and transformation of the spermatogenic cells in accordance with spermiogenesis. RELATION BETWEEN INFLAMMATORY CYTOKINES AND APOTOSIS EXPRESSION IN TENDON : AN ANALYSIS IN THE EQUINE SUPERHCIAL DIGITAL FLEXOR TENDON, Hosaka Yoshinao; Yamamoto Etsuko; Ueda Hiromi; Takehana Kazushige, Connective tissue, 34, (1) 86 - 86, 2002 , 【目的】アポトーシスは多くの組織で発現報告がなされているが、腱組織では明らかにされていない。本研究では腱組織での腱細胞のアポトーシス発現の有無を検索するとともに、正常時および炎症時での発現の差異を明らかにすることを目的とした。加えて、腱組織内での炎症性サイトカイン陽性細胞の分布を検索し、アポトーシス発現との関与を明らかにした。【材料と方法】ウマの浅指屈筋腱中央部を試料として用いた。試料は肉眼的に異常が全く認められなかったものを正常腱とし、出血・腫脹等の炎症像が見られたものを炎症腱とした。アポトーシス発現の検索はTUNEL法と抗Caspase3抗体による免疫組織化学的手法で、サイトカイン陽性細胞の分布は免疫組織化学的手法およびin situ hybridization法で検索した。形態変化については透過型電子顕微鏡で観察した。【結果と考察】正常腱、炎症腱ともにTUNEL陽性細胞およびCaspase3陽性細胞が確認された。正常腱では陽性細胞が散発的に分布していたのに対し、炎症腱では正常腱に比して多数の陽性細胞が分布していた。電顕でも核クロマチンの凝集像を持つ腱細胞が炎症腱で多く認められ、腱組織中でのアポトーシス発現が明らかになった。サイトカイン陽性細胞は正常腱では散発的に分布していたが、炎症腱では組織内に遊走したマクロファージ等の免疫細胞に加え、腱(芽)細胞、腱内膜細胞および血管内皮細胞が張陽性を呈した。腱炎時に腱組織の細胞外マトリックス構成成分が変化することはすでに明らかにされている。炎症による健内の環境変化が腱細胞のサイトカイン産生を促進し、産生されたサイトカインがさらに腱細胞のアポトーシスの誘発に関与していると推察した。 Liver fattening in cattle presenting with ketosis and its correlation with basal membrane inferior to sinusoid endothelial cell., 岸基史; 東伸好; 保坂善真; 植田弘美; 及川伸; 小岩政照; 竹花一成, 家畜診療, 49, (474) 771 - 776, 2002 Micromorphological characteristics of hepatic sinusoidal endothelial cells and their basal laminae in five different animal species, Nobuyo Higashi; Hiromi Ueda; Osamu Yamada; Shin Oikawa; Masateru Koiwa; Prasarn Tangkawattana; Kazushige Takehana, Okajimas Folia Anatomica Japonica, 79, (5) 135 - 142, 2002 , 10.2535/ofaj.79.135, The micromorphological characteristics of hepatic sinusoidal endothelial cells and their basal laminae in five different animal species (cow, sheep, guinea pig, pig, and dog) were studied by electron microscopy. Using transmission electron microscopy, a prominent basal lamina was seen only in cow and sheep. In pig and guinea pig, no prominent basal lamina was evident. Basal lamina-like material was occasionally found in dog. Immunohistochemically, basal lamina was found in cow, sheep, and dog. Using scanning electron microscopy, the size of endothelial fenestrae differed between species. Spleens from adult male rats were perfusion-fixed with 0.5% glutaraldehyde - 4 % paraformaldehyde containing 0.05% cetylpyridinium chloride and then perfused with 0.58 PEI at pH 7.4. On the free surfaces (glycocalyx of the plasma membrane) of the ECs examined, distinct PEI-positive reactions were observed in blood vessels, such as trabecular :arteries, central arteries, arterial capillaries, pulp veins and trabecular veins. These PEI-positive electron-dense substances in the trabecular arteries, central arteries, and trabecular veins took the shape of a band of 170-250 nm in thickness. On the other hand, the corresponding ultrastructure of the ECs lining the splenic sinuses and the RCs in the splenic cord showed exceedingly weak PEI reactions. The PEI-reactive deposits were significantly thinner than those in the above blood vessels. As the thickness of the electron-dense substances can be related to the density of the negative charge, these results suggest that there is a high intensity of negative charge on the free surfaces (glycocalyx of the plasma membrane) of ECs in blood vessels where blood cells and plasma pass into the red pulp ol are discharged from the red pulp. In contrast, the splenic sinuses and RCs, which are the main components of the red pulp, contain weakly negative-charged sites. This may contribute to the microcirculation of the splenic blood vessels and elucidate the possible physiological functions of the spleen, such as blood storage. The duodenal glands of the camel were distributed in the submucosa 2 m caudal from the pylorus. These were branched tubuloalveolar glands. The terminal portion of each lobule was formed by only one type of mucous cell. The duodenal gland cells contained acidic and neutral carbohydrates. The mucous cells mainly contained sulfate and carboxyl carbohydrate with sialic acid, and they also contained a few neutral carbohydrates with different saccharide residues such as mannose, glucose, galactose, N-acetyl glucosamine and N-acetyl galactosamine. The results showed that the secretary granules of the duodenal glands in the camel contain mainly acidic carbohydrates. These findings seem to be the morphological characteristics of the duodenal glands in the camel. The triple enzyme digestion method consists of a sequence of chondroitinase B, testicular hyaluronidase and heparitinase. The results obtained by the present experimental and control studies indicated that dermatan sulfates, chondroitin sulfates (A and/or C) or heparan sulfates were apparently observed in various ultrastructural features of aortic extracellular matrix, such as bundle of collagen fibers and soluble matrix of interstitial space. Particularly, we found that both heparan sulfates and chondroitin sulfates (A and/or C) were detected in association with the basal lamina of smooth muscle cells and the external surface of elastic lamina, and in the latter heparan sulfates were frequently recognized as a mass, whereas chondroitin sulfates (A and/or C) were found intermittently along the external surface of elastic lamina. This suggests that the triple enzyme digestion method which combines the glycosaminoglycans-degrading enzymes with the diamine reaction can be postulated to represent efficient and useful technique for precise electron microscopic histochemical detection of the glycosaminoglycans in the extracellular matrix of the rat aorta. The results indicated that the reticular cells contained an appreciable abundance of glycogen particles in the cytoplasm. In the free surface coat of the plasma membrane, the cells included neutral glycoprotein, which were devoid of detectable amounts of acidic glycoconjugates. The possible physiological significance of these carbohydrates is discussed, with special reference to their characteristic cytochemical features examined in the present study. Histochemical analysis of acidic glycoconjugates in the endothelium lining the splenic blood vessels in the rat, H Ueda; O Fujimori; M Abe, ARCHIVES OF HISTOLOGY AND CYTOLOGY, 59, (4) 389 - 397, 1996 10 , 10.1679/aohc.59.389, An electron microscopic analysis with specific histochemical stainings for acidic glycoconjugates was carried out to examine the endothelium lining blood vessels of the rat spleen, Histochemical staining performed was the postembedding high or low iron diamine-thiocarbohydrazide-silver protein-physical development (HID or LID-TCH-SP-PD) method, with or without prior digestion with acidic glycoconjugate-degrading enzymes, such as heparitinase, testicular hyaluronidase, chondroitinase B and neuraminidase. The results indicated that the acidic glycoconjugates in the basal lamina of the endothelial cells lining the four types of blood vessel(central arteries, arterial capillaries, splenic sinuses and pulp veins) were heparan sulfate, chondroitin sulfate A, and/or C, chondroitin sulfate B and sialic acid residues. In the endothelial cells lining the central arteries, arterial capillaries and pulp veins, the surface coat of the luminal plasma membrane included heparan sulfate, chondroitin sulfate A and/or C, chondroitin sulfate B and sialic acid residues, whereas the corresponding ultrastructure of the splenic sinuses was devoid of detectable amounts of acidic glycoconjugates. This suggests that such characteristic histochemical features of the endothelium in the four types of the splenic blood vessel can be related to the possible physiological functions of the spleen. Three types of mucous cells (types A-C) were differentiated on the basis of the reactivities of the secretory products elaborated by them. The carbohydrate contents of mucous globules predominantly comprised sulfate esters and traces of oxidizable vicinal diols in type-A cells, oxidizable vicinal diols in type-B cells, and moderate amounts of both sulfate esters and oxidizable vicinal diols in type-C cells. Glycogen particles were also found to occur in the cytoplasm of these cells and glycoproteins containing oxidizable vicinal diols were visualized in Golgi cisternae, rough endoplasmic reticulum, nuclear envelopes, and plasma membranes. In the type-A and type-B cells situated in the superficial layers of the epidermis, extensive cisternae of the Golgi apparatus and copious rough endoplasmic reticulum suggested the active syntheses of secretory contents, in contrast to the type-C mucous cells, which displayed poor development of these organelles, in the deeper layers. Pieces of the parotid, submandibular and sublingual glands from adult rats were subjected to glutaraldehyde-paraformaldehyde fixation, rinsed, dehydrated and embedded in LR-White resin. Ultrathin sections were prepared, grid-mounted, incubated in urea (6M) aqueous solution, and then stained with an alcian blue (AB) pH 1.0-phosphotungstic acid (PTA) sequence with or without prior treatment with nitrous acid. The results obtained have shown that sulfated glycoconjugates are involved in such ultrastructures as mast cell granules, basal lamina of arteriolar endothelial cells and intercellular matrix of arteriolar smooth muscle cells. CYTOPLASMIC FILAMENTS IN THE ENDOTHELIAL-CELLS OF THE SHEATHED CAPILLARY - AN ULTRASTRUCTURAL AND IMMUNOCYTOCHEMICAL STUDY IN THE PIG SPLEEN, K TAKEHANA; M ABE; M YAMAGUCHI; T UCHIDA; M INAGAKI; K YAMAMOTO; J MASTY; A WINNARD; H UEDA; H MIYATA, ACTA ANATOMICA, 143, (4) 294 - 300, 1992 04 , Cytoplasmic filaments of the endothelial cells of sheathed capillaries in the pig spleen were identified and their ultrastructure was studied. Two types of cytoplasmic filaments were found: intermediate filaments (diameter: 10 nm) which filled most of the interior of the cells, and thin filaments (diameter: 5 nm) which were located just beneath the cell membrane and filled the lateral cytoplasmic processes. In immunocytochemical preparations, the intermediate filaments were positive for vimentin and desmin, and were negative for keratin. Staining of the thin filaments with heavy meromyosin resulted in arrowhead formations. These observations suggest that the intermediate filaments maintain the cytoarchitecture, possibly protecting the cell from structural alterations induced by blood pressure changes. Concurrently, thin filaments may facilitate the passage of red blood cells and blood platelets through the interendothelial fenestrae of the sheathed endothelial cell to the reticular meshwork in the capillary sheath. ULTRASTRUCTURE OF THE RED PULP IN SPLEEN INNERVATION IN HORSE AND PIG, H UEDA; M ABE; K TAKEHANA; K IWASA; T HIRAGA, ACTA ANATOMICA, 141, (2) 151 - 158, 1991 06 , The innervation of the red pulp in the spleen of horse and pig was investigated by electron microscopy. In addition, the neurilemma was studied by immunohistochemistry specific for S-100 protein. In the pig, a large population of smooth-muscle cells extending from the smooth-muscle trabeculae was present in the red pulp. The cytoplasmic processes of reticular cells enwrapped the smooth-muscle cells, and nerve fibres were distributed between the smooth-muscle cells and the reticular cells. The nerve terminals clustered toward the facing of the smooth-muscle cells. Nerve fibres and terminals were not disclosed within the sheathed artery. Immunohistochemically the neurilemma showed a reaction positive for S-100 protein. In the horse, no smooth-muscle cells were noted in the red pulp. The nerve fibres terminated around the cytoplasmic processes of the reticular cells. Nerve fibres and terminals were disclosed within the sheathed artery, and the terminals contained both large and small dense-cored vesicles. Immunohistochemically the neurilemma showed a reaction negative for S-100 protein. These findings support the presence of the axon-bearing reticular cells described earlier in the horse spleen. It is a peracetic acid-thiocarbohydrazide-silver protein sequence followed by a physical development procedure. In the present study on paraffin and cryostat sections of liver, brain and ovary, unsaturated lipids were visualized as distinct reaction products coloured various shades of brown and black. The reaction products are easier to see and the method is more efficient than the peracetic acid-Schiff method. Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Cloning and functional analysis of taste receptors for sourness and saltiness, 1998, 2000, SHIMADA Shoichi; UEDA Takashi; HIRABAYASHI Yoshifumi; FUJIMORI Osamu; UGAWA Shinya, We have identifieda cDNA encoding a taste receptor for sourness (MDEG1) from a rat circumvallate papilla cDNA library by combined cDNA homology and functional expression approaches. When expressed in Xenopus laevis oocytes, MDEG1 showed characteristics of a proton-gated amiloride-sensitive cation channel. Interestingly, acidic stimulation by acetic acid, the chief ingredient of vinegar, generatedlarger inwardcurrents than that by hydrochloric acidat equal pH.In situ hybridization revealed that the mRNA of MDEG1 was exclusively localized in taste buds. MDEG1 immunoreactivity was found on the cell surface and in the apical portion of taste bud cells, especially in type III cells which are classifiedas gustatory receptor cells. This is the first taste receptor cDNA to be cloned. To isolate another subunit of the sour-taste receptor, we screened a rat circumvallate papilla cDNA library and iden tified MDEG2. Immunohistochemical and electrophysiological studies demonstrated that MDEG2 formed heterooligomeric channels with MDIEG1 at the apical portion of single taste-cells and modified the desensitization of MDEG1 channel. The desensitization rate of the heterooligomer was dramatically altered by a single amino acid substitution of Gly481 in MDEG2 without changing other channel properties. Active Expression of Matrix Metalloproteinase-13 mRNA in the Granulation Tissue of Equine Superficial Digital Flexor Tendinitis, NOMURA Masayuki; HOSAKA Yoshinao; KASASHIMA Yoshinori; UEDA Hiromi; TAKEHANA Kazushige; KUWANO Atsutoshi; ARAI Katsuhiko, Journal of Veterinary Medical Science, 69, (6) 637 - 639, 2007 06 25 , The DNA microarray analysis for matrix metalloproteinase (MMP)-related mRNA expression in equine superficial digital flexor tendinitis indicated that mRNA level of MMP-13 was apparently up-regulated in the tendinitis as compared to normal tendon. <i>In situ</i> hybridization also revealed that fibroblastic cells proliferated in the granulation tissue generated in the tendinitis actively expressed MMP-13 mRNA. On the other hand, in normal tendon, a few fibroblastic cells and vascular components lied in the endotenon barely expressed its mRNA, but other cellular components in the tendon bundle were not positively hybridized. As mentioned above, MMP-13 but not other collagenases or gelatinases, may play an important role in tendon injuries in the racehorses.<br> |

Tendon tissues for tissue culture were isolated from Achilles tendons of rabbits. The tendon segments were placed one segment per well and incubated in growth medium consisting of Dullbecco's modified Eagle's medium supplemented with 5% fetal bovine serum at 37 degrees C in a humidified atmosphere with 5% CO2 for various periods. The alignment of collagen fibrils was preserved for 48 h, but tendon structure has disintegrated at 96 h. Alcian blue staining and gelatine zymography revealed that proteoglycan markedly diminished and that matrix metalloproteinase (MMPs) activity was upregulated sharply at 72 and 96 h. The ratio of collagen fibrils with large diameter had increased and the mean diameter and mass average diameter value had reached maximum at 48 h. The values then decreased and mean diameters at 72 and 96 h were significantly different from that at 48 h. At 96 h, the ratio of collagen fibrils with small diameters had increased and collagen fibrils with large diameters had disappeared. These findings indicate that structural alteration is possible to be induced by disintegration of collagen fibrils and disappearance of glycosaminoglycans from extracellular matrix (ECM), subsequent of upregulation of MMPs activity. Although the study period is limited, the tissue culture method is available for investigating cell-ECM interaction in tendons. |

The Molecular Marker NTx Reflecting Changes in Tissue Remodeling Associated with Injury to the Equine Superficial Digital Flexor Tendon, HOSAKA Yoshinao; HIGUCHI Hidetoshi; ONAGA Takenori; NAGATA Hiroshi; KAMIKURA Tomoki; MISUMI Kazuhiro; UEDA Hiromi; Kazushige, Journal of the Japan Veterinary Medical Association, 58, (3) 175 - 179, 2005 03 20 , 骨の代謝マーカーの一つであるI型コラーゲン架橋N-末端テロペプチド濃度(Nx)が馬の腱損傷後の治癒過程をモニタリングするマーカーとして有用であるかを検討した。血清中Nx濃度は腱損傷のない正常群と比較して、腱炎の急性期および亜急性期群で有意に高い値を示した。しかし、慢性期群と正常群間とのNx値には有意差が認められなかった。腱炎による腱のI型コラーゲンの分解は亜急性期までで、慢性期以降では腱組織の再構築が開始されることから、血清中Nx濃度は腱炎によるコラーゲン分解を鋭敏に反映していると考えられた。 |

Effects of ingestion of collagen peptide on collagen fibrils and glycosaminoglycans in Achilles tendon, Jun Minaguchi; Yoh-Ichi Koyama; Natsuko Meguri; Yoshinao Hosaka; Hiromi Ueda; Masashi Kusubata; Arisa Hirota; Shinkichi Irie; Naoki Mafune; Kazushige Takehana, Journal of Nutritional Science and Vitaminology, 51, (3) 169 - 174, 2005 , 10.3177/jnsv.51.169, In order to investigate whether the oral ingestion of collagen peptide affects the extracellular matrix of tendon, two doses (0.2 g/kg and 1.0 g/kg body weight) were orally administered daily for 56 d to a rabbit, and both the size of collagen fibrils and the amount of glycosaminoglycans in the Achilles tendon were measured in comparison with those in a rabbit fed with a control protein, lactalbumin, or water alone. Ingestion of collagen peptide or lactalbumin induced a significant increase in collagen fibril diameter and a decrease in fibril density except for a high dose of lactalbumin compared with the water control. A histogram pattern of fibril diameter in a high dose of collagen peptide showed a peak at 160-180 nm, which was not observed in other groups. However the percentage of diameters over 200 nm was the lowest in this group but highest in the low-dose group of collagen peptide. The mean fibril diameter and mass average diameter of a high dose of collagen peptide were significantly smaller than those in a low dose. The amount of dermatan sulphate increased in the high-dose groups, while the amount of hyaluronic acid decreased in rabbits fed with collagen peptide or lactalbumin at either dose. These results suggest that the ingestion of collagen peptide affects the size of collagen fibrils and composition of glycosaminoglycans in the Achilles tendon and thus may improve the mechanical properties of the Achilles tendon. |
Mechanism of cell death in inflamed superficial digital flexor tendon in the horse, Y Hosaka; H Teraoka; E Yamamoto; H Ueda; K Takehana, JOURNAL OF COMPARATIVE PATHOLOGY, 132, (1) 51 - 58, 2005 01 , 10.1016/j.jcpa.2004.06.006, The aim of the present study was to clarify the presence and determine the role of apoptosis in the degenerative process of the superficial digital flexor tendon (SDFT) in the horse. Samples were obtained from normal and inflamed SDFTs of horses. To detect apoptosis and to identify apoptotic cells, the samples were subjected to immunohistochemical labelling and Western blot analysis. Although a large number of cells in degenerate areas showed positive reactions with caspase-3 and single stranded DNA antibodies, cells in normal tendon samples showed very weak reactions. Excessive apoptosis was confirmed by the results of Western blot analysis, which showed a significant increase in activated caspase-3 protein in the inflamed SDFTs, suggesting that apoptosis occurred in the tendinocytes via a caspase-3-dependent pathways This is the first report of excessive apoptosis in inflamed SDFT of the horse. The results indicate that apoptosis may play an important role in the process of degeneration of the tendon as well as other tissues. (C) 2004 Elsevier Ltd. All rights reserved. |

Differences in Tumor Necrosis Factor (TNF)α and TNF Receptor-1-Mediated Intracellular Signaling Factors in Normal, Inflamed and Scar-Formed Horse Tendons, HOSAKA Yoshinao; KIRISAWA Rikio; UEDA Hiromi; YAMAGUCHI Mamoru; TAKEHANA Kazushige, Journal of Veterinary Medical Science, 67, (10) 985 - 991, 2005 , 10.1292/jvms.67.985, Tumor necrosis factor (TNF) receptors (TNF-R)-mediated cell survival or apoptosis has been demonstrated in many cells, but little is known about survival or apoptotic signals via TNF-R1 in tendinocytes. In this study, we focused on four signaling factors, TNFα, TNF-R1, TNFR-associated factor2 (TRAF2) and caspase-3, in order to elucidate the signaling events in tendinocytes. Samples were obtained from normal, inflamed and scar-formed equine superficial digital flexor tendons. To detect these signaling factors, samples were subjected to immunohistochemistry and Western blot analysis, and some samples were also subjected to reverse transcription-polymerase chain reaction (RT-PCR), PCR-Southern blot analysis and in situ hybridization to detect the expression of TNFα mRNA. Distribution of the four factors differed depending on the tendon condition, normal, inflamed or scar-formed. In the normal tendon, large amounts of TRAF2 were found in tendinocytes, but the amounts of TNF-R1 were small. TNFα mRNA was expressed most highly in the inflamed tendon. TNF-R1, which was only faintly detected in the normal tendon, was detected at a high level in the inflamed tendon, and the amounts of TRAF2 and caspase-3 also increased. Activated caspase-3 was only detected in the inflamed tendon. TNFα mRNA was also expressed in the scar-formed tendon, though it showed weak signals, and the expression levels of TNF-R1, TRAF2 and caspase-3 proteins were very low. Two distinct intracellular signaling pathways of TNFα, which lead to cell survival and apoptosis, might be present in tendinocytes mediated through TNF-R1. These results, which reflect the dynamism of TNFα, provide important clues for means to prevent tendinopathy. |

Type III collagen content increased with aging. In terms of glycosaminoglcan composition, dermatan sulfate decreased and chondroitin sulfate increased. In the SDFT of thoroughbred horses, morphological age-related changes in collagen fibrils are accompanied by changes in biochemical composition. |

In this study, the localizations of 4 cytokines (IL-1alpha, IL-1beta, TNFalpha and IFNgamma) in tendinocytes of the equine superficial digital flexor tendon (SDFT) were analyzed by the use of an immunohistochemical method. In inflamed tendons positive staining for all 4 cytokines antibodies were detected in endotedinieum cells and vascular epithelial cells. In contrast, negative or trace immunoreactions were obtained in many tendinocytes in the normal tendon. The variation in cellular immune responses depending on the kind of cytokine may reflect the physiological/pathological condition of the SDFT. |

イヌの角膜成熟と細胞外マトリックス成分の相関性, 竹花一成; 山本悦子; 保坂善真; 植田弘美, マトリックス研究会大会, 49th, (1) 75 - 75, 2002 04 , 【目的】新生イヌの角膜は未発達な網膜や輝板を保護するために混濁しており、眼の成熟と共に透明になる.角膜の透明度は角膜固有層のコラーゲン細線維の規則正しい配列とそれらの間の細胞外マトリックスの一種であるグリコサミノグリカン(GAG)の充填が重要であるとされている.しかし,混濁とGAG構成の変化との相関性については明らかにされていない.そこでイヌの角膜の正常変化についてビーグル犬を用い形態学的および生化学的手法により解析した.【方法】形態学的には角膜のコラーゲン細線維の配列状態ならびに直径と単位面積あたりの基質占有割合を算出し,生化学的にはGAGの解析を行った.【結果】新生イヌの角膜は開眼時には混濁しており,その後徐々に混濁は和らぎ,生後3週目前後で完全に透明になった.角膜の主体である角膜固有層のコラーゲン細線維の直径は週齢毎でほぼ均一で0週齢では24.0nm程度,成イヌでは38.0nmであった.コラーゲン細線維の配列は成熟に伴い規則性を有していた.基質占有割合は80%から30%と減少していた.基質のGAG組成は週齢の増加と共にケラタン硫酸(KS)が増加しデルマタン硫酸(DS)が減少していた.【考察】角膜成熟に伴いコラーゲン細線維の太さと規則性の変化が認められ、生化学的には、KSの増加とDSの減少が顕著であった。このことは、コラーゲン細線維に対するKSとDSの作用、すなわちその直径と空間の維持と形成抑制作用に由来するもので、角膜の成熟にはこれらの成分の存在は必要不可欠であることが明らかになった。 |
けん成熟に対するMMPの作用, 山本悦子; 保坂善真; 植田弘美; 竹花一成, マトリックス研究会大会, 49th, (1) 88 - 88, 2002 04 , 【目的】浅指屈筋腱は腱細胞、コラーゲン細線維と非コラーゲン性マトリックスにより構成されている。ウマの腱における細胞外マトリックスの加齢性変化の研究は、コラーゲンについては多数報告されているが、マトリックス分解系についての報告はほとんどない。そこで、コラーゲンの細胞外分傍系の1つであるマトリツクスメタロブロテアーゼ(MMP)に著目し、腱の加齢性変化とMMPの関与を検索した。【材料および方法】材料は、サラブレッド種(0歳5例、1歳2例、2歳2例、3歳1例、計10例)の浅指屈筋腱中手骨中央部を用いた。形態学的には、電子顕微鏡を用いてコラーゲン細線維の走行と直径の変化を視察した。生化学的には、二次元電気泳動法にてグリコサミノグリカン(GAG)の分離・同定を行った。さらに、Film in situ ZymographyとザイモグラフィーにてMMPの分離・同定を行った。【結果および考察】コラーゲン細線維は0歳では波状に走行しており、3歳では長軸に沿って平行に走行していた。細線維直径は0歳では20nmと100nmが多く、加齢に伴ってそれぞれ60nm、220nmと太くなり、その割合は加齢に伴い細いものが増加した。GAGはヒアルロン酸、コンドロイチン硫酸、デルマタン硫酸で構成されており、加齢に伴いデルマタン硫酸が減少し、コンドロイチン硫酸が増加した。MMPはMMP-2とMMP-9の潜在型と活性型が存在し、加齢に伴いMMP-9が流少し、MMP-2が増加した。以上のことより、腱コラーゲン細線維の加齢に伴う形態変化は、コラーゲン細線維形成調節作用を持つGAGによって起こり、分解系であるMMPも作用していることが示唆された。 |

Ultrastructure and function of sertoli cell processes in the Korean native goat, Geon Oh Kwon; Young Seok Park; Seong Ho Lee; Etsuko Yamamoto; Yoshinao Hosaka; Hiromi Ueda; Kazushige Takehana, Okajimas Folia Anatomica Japonica, 79, (4) 121 - 128, 2002 , 10.2535/ofaj.79.121, The infrastructure of Sertoli cell in the testes of Korean native goats was investigated by scanning and transmission electron microscopy to elucidate the relationship between germ cell and Sertoli cell processes in the spermiogenetic cycle. Type-A Sertoli cells at stage V of the spermiogenetic cycle and type-B Sertoli cells at stage Vu of the spermiogenetic cycle were used for analysis. Morphologically, the Sertoli cell processes were classified into sheet-like and slender cord-like processes. The sheet-like process originated solely from the Sertoli cell column while the slender cord-like process projected either from the Sertoli cell column or the sheet-like process. Periodic acid (PA)-thiocarbohydrazide (TCH)-silver protein (SP)-physical development (PD)-positive granules were found diffusively both in the sheet-like and slender cord-like processes near the round spermatid, whereas they had accumulated near the head of the elongated spermatid. The morphological variation and glucoconjugate histochemical reaction of the Sertoli cell processes reflect nourishment, movement and transformation of the spermatogenic cells in accordance with spermiogenesis. |
RELATION BETWEEN INFLAMMATORY CYTOKINES AND APOTOSIS EXPRESSION IN TENDON : AN ANALYSIS IN THE EQUINE SUPERHCIAL DIGITAL FLEXOR TENDON, Hosaka Yoshinao; Yamamoto Etsuko; Ueda Hiromi; Takehana Kazushige, Connective tissue, 34, (1) 86 - 86, 2002 , 【目的】アポトーシスは多くの組織で発現報告がなされているが、腱組織では明らかにされていない。本研究では腱組織での腱細胞のアポトーシス発現の有無を検索するとともに、正常時および炎症時での発現の差異を明らかにすることを目的とした。加えて、腱組織内での炎症性サイトカイン陽性細胞の分布を検索し、アポトーシス発現との関与を明らかにした。【材料と方法】ウマの浅指屈筋腱中央部を試料として用いた。試料は肉眼的に異常が全く認められなかったものを正常腱とし、出血・腫脹等の炎症像が見られたものを炎症腱とした。アポトーシス発現の検索はTUNEL法と抗Caspase3抗体による免疫組織化学的手法で、サイトカイン陽性細胞の分布は免疫組織化学的手法およびin situ hybridization法で検索した。形態変化については透過型電子顕微鏡で観察した。【結果と考察】正常腱、炎症腱ともにTUNEL陽性細胞およびCaspase3陽性細胞が確認された。正常腱では陽性細胞が散発的に分布していたのに対し、炎症腱では正常腱に比して多数の陽性細胞が分布していた。電顕でも核クロマチンの凝集像を持つ腱細胞が炎症腱で多く認められ、腱組織中でのアポトーシス発現が明らかになった。サイトカイン陽性細胞は正常腱では散発的に分布していたが、炎症腱では組織内に遊走したマクロファージ等の免疫細胞に加え、腱(芽)細胞、腱内膜細胞および血管内皮細胞が張陽性を呈した。腱炎時に腱組織の細胞外マトリックス構成成分が変化することはすでに明らかにされている。炎症による健内の環境変化が腱細胞のサイトカイン産生を促進し、産生されたサイトカインがさらに腱細胞のアポトーシスの誘発に関与していると推察した。 |

Using transmission electron microscopy, a prominent basal lamina was seen only in cow and sheep. In pig and guinea pig, no prominent basal lamina was evident. Basal lamina-like material was occasionally found in dog. Immunohistochemically, basal lamina was found in cow, sheep, and dog. Using scanning electron microscopy, the size of endothelial fenestrae differed between species. These results may suggest that variation of endothelia and their basal laminae of different species is related to differences in nourishment. |

Electron microscopic cytochemical studies of anionic sites in the rat spleen, H Ueda; K Takehana; Eerdunchaolu; K Iwasa; O Fujimori; S Shimada, JOURNAL OF VETERINARY MEDICAL SCIENCE, 63, (3) 287 - 291, 2001 03 , 10.1292/jvms.63.287, The distribution patterns of the intensity of negative charge on the free surfaces (glycocalyx of the plasma membrane) of endothelial cells (ECs) in blood vessels and reticular cells (RCs) in the splenic cord of the rat spleen were studied by an electron microscopic cytochemical method using polyethyleneimine (PEI) as a cationic probe. Spleens from adult male rats were perfusion-fixed with 0.5% glutaraldehyde - 4 % paraformaldehyde containing 0.05% cetylpyridinium chloride and then perfused with 0.58 PEI at pH 7.4. On the free surfaces (glycocalyx of the plasma membrane) of the ECs examined, distinct PEI-positive reactions were observed in blood vessels, such as trabecular :arteries, central arteries, arterial capillaries, pulp veins and trabecular veins. These PEI-positive electron-dense substances in the trabecular arteries, central arteries, and trabecular veins took the shape of a band of 170-250 nm in thickness. On the other hand, the corresponding ultrastructure of the ECs lining the splenic sinuses and the RCs in the splenic cord showed exceedingly weak PEI reactions. The PEI-reactive deposits were significantly thinner than those in the above blood vessels. As the thickness of the electron-dense substances can be related to the density of the negative charge, these results suggest that there is a high intensity of negative charge on the free surfaces (glycocalyx of the plasma membrane) of ECs in blood vessels where blood cells and plasma pass into the red pulp ol are discharged from the red pulp. In contrast, the splenic sinuses and RCs, which are the main components of the red pulp, contain weakly negative-charged sites. This may contribute to the microcirculation of the splenic blood vessels and elucidate the possible physiological functions of the spleen, such as blood storage. |

The duodenal glands of the camel were distributed in the submucosa 2 m caudal from the pylorus. These were branched tubuloalveolar glands. The terminal portion of each lobule was formed by only one type of mucous cell. The duodenal gland cells contained acidic and neutral carbohydrates. The mucous cells mainly contained sulfate and carboxyl carbohydrate with sialic acid, and they also contained a few neutral carbohydrates with different saccharide residues such as mannose, glucose, galactose, N-acetyl glucosamine and N-acetyl galactosamine. The results showed that the secretary granules of the duodenal glands in the camel contain mainly acidic carbohydrates. These findings seem to be the morphological characteristics of the duodenal glands in the camel. |

Effects of a triple enzyme digestion method on a diamine reaction for glycosaminoglycans of the at aorta in electron microscopy, H Ueda; O Fujimori, JOURNAL OF VETERINARY MEDICAL SCIENCE, 60, (11) 1169 - 1174, 1998 11 , 10.1292/jvms.60.1169, Effects of a triple enzyme digestion method using glycosaminoglycans-degrading enzymes upon a diamine reaction have been tested for electron microscopic histochemical detection of glycosaminoglycans in extracellular matrix of the rat aorta. The triple enzyme digestion method consists of a sequence of chondroitinase B, testicular hyaluronidase and heparitinase. The results obtained by the present experimental and control studies indicated that dermatan sulfates, chondroitin sulfates (A and/or C) or heparan sulfates were apparently observed in various ultrastructural features of aortic extracellular matrix, such as bundle of collagen fibers and soluble matrix of interstitial space. Particularly, we found that both heparan sulfates and chondroitin sulfates (A and/or C) were detected in association with the basal lamina of smooth muscle cells and the external surface of elastic lamina, and in the latter heparan sulfates were frequently recognized as a mass, whereas chondroitin sulfates (A and/or C) were found intermittently along the external surface of elastic lamina. This suggests that the triple enzyme digestion method which combines the glycosaminoglycans-degrading enzymes with the diamine reaction can be postulated to represent efficient and useful technique for precise electron microscopic histochemical detection of the glycosaminoglycans in the extracellular matrix of the rat aorta. |
Cytochemical studies on carbohydrates in the reticular cells of the rat splenic cord, H Ueda; O Fujimori, ACTA HISTOCHEMICA ET CYTOCHEMICA, 31, (1) 77 - 79, 1998 , 10.1267/ahc.31.77, Carbohydrates in the reticular cells of the rat splenic cord were studied by means of a series of electron microscopic cytochemical methods. The results indicated that the reticular cells contained an appreciable abundance of glycogen particles in the cytoplasm. In the free surface coat of the plasma membrane, the cells included neutral glycoprotein, which were devoid of detectable amounts of acidic glycoconjugates. The possible physiological significance of these carbohydrates is discussed, with special reference to their characteristic cytochemical features examined in the present study. |

The results indicated that the acidic glycoconjugates in the basal lamina of the endothelial cells lining the four types of blood vessel(central arteries, arterial capillaries, splenic sinuses and pulp veins) were heparan sulfate, chondroitin sulfate A, and/or C, chondroitin sulfate B and sialic acid residues. In the endothelial cells lining the central arteries, arterial capillaries and pulp veins, the surface coat of the luminal plasma membrane included heparan sulfate, chondroitin sulfate A and/or C, chondroitin sulfate B and sialic acid residues, whereas the corresponding ultrastructure of the splenic sinuses was devoid of detectable amounts of acidic glycoconjugates. This suggests that such characteristic histochemical features of the endothelium in the four types of the splenic blood vessel can be related to the possible physiological functions of the spleen. |

Three types of mucous cells (types A-C) were differentiated on the basis of the reactivities of the secretory products elaborated by them. The carbohydrate contents of mucous globules predominantly comprised sulfate esters and traces of oxidizable vicinal diols in type-A cells, oxidizable vicinal diols in type-B cells, and moderate amounts of both sulfate esters and oxidizable vicinal diols in type-C cells. Glycogen particles were also found to occur in the cytoplasm of these cells and glycoproteins containing oxidizable vicinal diols were visualized in Golgi cisternae, rough endoplasmic reticulum, nuclear envelopes, and plasma membranes. In the type-A and type-B cells situated in the superficial layers of the epidermis, extensive cisternae of the Golgi apparatus and copious rough endoplasmic reticulum suggested the active syntheses of secretory contents, in contrast to the type-C mucous cells, which displayed poor development of these organelles, in the deeper layers. |
A POSTEMBEDDING METHOD USING UREA-UNMASKED ULTRATHIN SECTIONS FOR THE ELECTRON-MICROSCOPIC DETECTION OF SULFATED GLYCOCONJUGATES, H UEDA; O FUJIMORI; K YAMADA, ACTA HISTOCHEMICA ET CYTOCHEMICA, 26, (1) 27 - 34, 1993 , 10.1267/ahc.26.27, Using urea-unmasked ultrathin sections, a postembedding staining method in combination with nitrous acid procedure has been established for the efficient visualization of sulfated glycoconjugates in a series of ultrastructures of three salivary glands of the rat. Pieces of the parotid, submandibular and sublingual glands from adult rats were subjected to glutaraldehyde-paraformaldehyde fixation, rinsed, dehydrated and embedded in LR-White resin. Ultrathin sections were prepared, grid-mounted, incubated in urea (6M) aqueous solution, and then stained with an alcian blue (AB) pH 1.0-phosphotungstic acid (PTA) sequence with or without prior treatment with nitrous acid. The results obtained have shown that sulfated glycoconjugates are involved in such ultrastructures as mast cell granules, basal lamina of arteriolar endothelial cells and intercellular matrix of arteriolar smooth muscle cells. |
CYTOPLASMIC FILAMENTS IN THE ENDOTHELIAL-CELLS OF THE SHEATHED CAPILLARY - AN ULTRASTRUCTURAL AND IMMUNOCYTOCHEMICAL STUDY IN THE PIG SPLEEN, K TAKEHANA; M ABE; M YAMAGUCHI; T UCHIDA; M INAGAKI; K YAMAMOTO; J MASTY; A WINNARD; H UEDA; H MIYATA, ACTA ANATOMICA, 143, (4) 294 - 300, 1992 04 , Cytoplasmic filaments of the endothelial cells of sheathed capillaries in the pig spleen were identified and their ultrastructure was studied. Two types of cytoplasmic filaments were found: intermediate filaments (diameter: 10 nm) which filled most of the interior of the cells, and thin filaments (diameter: 5 nm) which were located just beneath the cell membrane and filled the lateral cytoplasmic processes. In immunocytochemical preparations, the intermediate filaments were positive for vimentin and desmin, and were negative for keratin. Staining of the thin filaments with heavy meromyosin resulted in arrowhead formations. These observations suggest that the intermediate filaments maintain the cytoarchitecture, possibly protecting the cell from structural alterations induced by blood pressure changes. Concurrently, thin filaments may facilitate the passage of red blood cells and blood platelets through the interendothelial fenestrae of the sheathed endothelial cell to the reticular meshwork in the capillary sheath. |
ULTRASTRUCTURE OF THE RED PULP IN SPLEEN INNERVATION IN HORSE AND PIG, H UEDA; M ABE; K TAKEHANA; K IWASA; T HIRAGA, ACTA ANATOMICA, 141, (2) 151 - 158, 1991 06 , The innervation of the red pulp in the spleen of horse and pig was investigated by electron microscopy. In addition, the neurilemma was studied by immunohistochemistry specific for S-100 protein. In the pig, a large population of smooth-muscle cells extending from the smooth-muscle trabeculae was present in the red pulp. The cytoplasmic processes of reticular cells enwrapped the smooth-muscle cells, and nerve fibres were distributed between the smooth-muscle cells and the reticular cells. The nerve terminals clustered toward the facing of the smooth-muscle cells. Nerve fibres and terminals were not disclosed within the sheathed artery. Immunohistochemically the neurilemma showed a reaction positive for S-100 protein. In the horse, no smooth-muscle cells were noted in the red pulp. The nerve fibres terminated around the cytoplasmic processes of the reticular cells. Nerve fibres and terminals were disclosed within the sheathed artery, and the terminals contained both large and small dense-cored vesicles. Immunohistochemically the neurilemma showed a reaction negative for S-100 protein. These findings support the presence of the axon-bearing reticular cells described earlier in the horse spleen. |

It is a peracetic acid-thiocarbohydrazide-silver protein sequence followed by a physical development procedure. In the present study on paraffin and cryostat sections of liver, brain and ovary, unsaturated lipids were visualized as distinct reaction products coloured various shades of brown and black. The reaction products are easier to see and the method is more efficient than the peracetic acid-Schiff method. |

Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Cloning and functional analysis of taste receptors for sourness and saltiness, 1998, 2000, SHIMADA Shoichi; UEDA Takashi; HIRABAYASHI Yoshifumi; FUJIMORI Osamu; UGAWA Shinya, We have identifieda cDNA encoding a taste receptor for sourness (MDEG1) from a rat circumvallate papilla cDNA library by combined cDNA homology and functional expression approaches. When expressed in Xenopus laevis oocytes, MDEG1 showed characteristics of a proton-gated amiloride-sensitive cation channel. Interestingly, acidic stimulation by acetic acid, the chief ingredient of vinegar, generatedlarger inwardcurrents than that by hydrochloric acidat equal pH.In situ hybridization revealed that the mRNA of MDEG1 was exclusively localized in taste buds. MDEG1 immunoreactivity was found on the cell surface and in the apical portion of taste bud cells, especially in type III cells which are classifiedas gustatory receptor cells. This is the first taste receptor cDNA to be cloned. To isolate another subunit of the sour-taste receptor, we screened a rat circumvallate papilla cDNA library and iden tified MDEG2. Immunohistochemical and electrophysiological studies demonstrated that MDEG2 formed heterooligomeric channels with MDIEG1 at the apical portion of single taste-cells and modified the desensitization of MDEG1 channel. The desensitization rate of the heterooligomer was dramatically altered by a single amino acid substitution of Gly481 in MDEG2 without changing other channel properties. These findings suggest that MDEG2 regulates the sour-taste receptor desensitization contributing to sour-taste adaptation and that Gly481 in MDEG2 is the key amino acidresidue that controls the duration of sourtaste sensation. |
