Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research Grant-in-Aid for Research Activity start-up, Elucidation of the DGK regulated erythrocyte invasion mechanism of malaria parasites, 202009, 202203, Takahiro Ishizaki, マラリア原虫は赤血球寄生原虫であり、宿主赤血球へと侵入する際に細胞内シグナル伝達を介して原虫タンパク質の分泌を制御する。ネズミマラリア原虫(Plasmodium yoelii)が保有するジアシルグリセロール(DAG)をホスファチジン酸(PA)へと転換させるジアシルグリセロールキナーゼ(DGK)の1つであるDGK1欠損マラリア原虫は赤血球侵入関連分子の細胞内局在の変化、分子分泌の低下、その結果に伴う原虫のマウスへの感染率と病原性の低下という表現型を示した。ネズミマラリア原虫は2つのDGKを有しており、その2つが相補的な機能を有しているかを検討するためにDGK1/DGK3二重欠損原虫を作出した。DGK1欠損原虫とDGK1/DGK3二重欠損原虫間における赤血球侵入効率には変化がなかったため、DGK1のみがマラリア原虫の赤血球侵入時に必要な酵素であり、赤血球侵入過程において2つのDGK間に相補的作用はないという結論に至った。 またDGK1によって転換されるPAはPHドメインを有するタンパク質と結合することが知られており、原虫が保有するPHドメイン誘導型欠損原虫の作出と解析を並行して実施した。その結果PHドメインを有する分子の1つであるAPHを誘導型欠損した原虫でDGK1欠損原虫同様に赤血球侵入関連分子の表面分泌が阻害される知見を得た。その一方でAPH欠損原虫では赤血球侵入関連分子の細胞内局在の変化は見られなかった。これらの結果からDGK1によって転換されるPAとAPHの相互作用が赤血球侵入関連分子の分泌には必須だが、それら分子の細胞内局在はDGK1由来のPAのみが関与する可能性が示唆された。

Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research Grant-in-Aid for JSPS Fellows, マラリア原虫の赤血球侵入関連分子分泌調節カスケードの解明, 20170426, 20200331, 石崎 隆弘, マラリア原虫の赤血球侵入は原虫の生存に必須なステップであり、宿主への病原性と密接な繋がりを持つ。赤血球から脱出した原虫はマイクロネームやロプトリーといった細胞小器官から種々の分子を順序立てて分泌しながら赤血球侵入を行う。マイクロネーム分子の分泌には細胞外液組成である低カリウム条件に起因するCa2+シグナルが関与することが明らかとなっているが、分子分泌に至るシグナル経路には不明な点が多い。本研究はネズミマラリア原虫を用いて、後期シゾント期における発現が高いキナーゼ群をDiCre-loxPシステムにより誘導型欠損させた原虫を使って、赤血球侵入に関わる標的キナーゼの同並びに、表現系の得られた分子の下流分子の探索を目的とする。 本年度は、得られたloxP挿入原虫もしくは遺伝子欠損原虫のうち、3つのキナーゼにおいて侵入能力が有意に減少するという結果が得られた。これら得られた3つのキナーゼのうち、2つのキナーゼは侵入能力、マウスへの病原性に対して顕著な減少が見られたことから赤血球侵入関連分子の分泌を免疫蛍光抗体法により評価した。その結果1つのキナーゼでは分子分泌のタイミングが遅れるということ、別のキナーゼでは侵入関連分子群の分泌そのものが低下するということが明らかとなった。これらの結果が引き起こされる分子メカニズムを明らかにするために、次年度はリン酸化プロテオーム解析及び、リン脂質メディエーターの探索を行う予定である。
