The cow laid down, lactation decreased, lost its appetite, and the prognosis was poor. No gross abnormalities were detected during the necropsy. Histopathological examination of the cerebrum, diencephalon, midbrain, pons, and medulla oblongata revealed multiple vessels cuffed by numerous lymphocytes and glial nodules, which were more severe in the brain stem. No pathogenic bacteria were isolated from the cerebrum and other organs, and no other viruses were detected. The nucleotide sequence of the detected AstV gene fragment had a 95% coincidence with the non-structural protein region of neurotropic AstV reported in the first confirmed case in Japan. AstV may have been involved in this case of encephalitis, which is considered to be the second reported case in Japan. This study suggests the existence of neurotropic AstV in multiple regions of Japan and infers the possibility that AstV is prevalent in Japan.

プリオン病は進行性の脳神経細胞変性を起こす一群のまれな致死性疾患で、代表的な疾患としては人のクロイツフェルト・ヤコブ病(CJD)や牛海綿状脳症(狂牛病、BSE)がある。BSEは人に伝達することが分かっている(変異型CJD)。この疾患の原因は細胞内の異常なプリオンタンパク(PrPSc)の蓄積によるものと考えられているが、どのような機序で発症するのか詳しく分かっていない。現在のところ有効な治療法もない。発症機序の解明や治療法の開発には動物モデルが必要であり、齧歯類等を用いたプリオン病モデルは作出されているが、これらのモデルでは人と同様な高次脳機能障害を再現することは難しい。従って、人に近い霊長類を用いたプリオン病モデルは必要とされる。BSEを発症したウシの脳乳剤を霊長類に接種し、人と同様の症状を呈することは既に報告されている。しかしながら、脳内接種し発症するまでに2 年以上、発症期間は1年前後を要する。 また個体による潜伏期間及び発症期間の差が大きい。 そこで、本研究では、より安定的なモデルの構築を目的に、PrPSc 持続発現細胞株を3頭のカニクイザル脳内に接種した。その結果、全頭とも接種1年4,5ヶ月後に神経症状のひとつである驚愕反応等を示し、その後急速に症状が悪化した。安楽死の目安とした神経症状スコアに達したため、接種1年7ヶ月後に全頭、安楽死を行った。安楽死直後のMRI撮像により、プリオン病に見られる脳の萎縮、顕著な脳室拡張が認められた。また病理検索やウエスタンブロッティングにより脳内のPrPSc蓄積も確認した。PrPSc細胞株を用いることにより、従来モデルに比べ、潜伏期間や症状の進行が早く、且つ個体による発症経過の差が少ない安定したモデルが構築できた。実験期間の短縮は感染ザルの負担軽減にも寄与する。今後はこのモデルを利用して、プリオン病の発症機序の解明や治療法の開発等を試みる。

道内で特用家畜・家禽として飼育されるアルパカ(Vicugna pacos)とエミュー(Dromaius novaehollandiae)の寄生虫保有状況に関する予備試験, 高野結衣; 竹内萌香; 立本完吾; 萩原克郎; 浅川満彦, 北海道獣医師会雑誌, 60, (9) 427 - 429,   2016 08 , 道内の観光牧場などで飼育されていた特用家畜アルパカ(Vicugna pacos)と特用家禽エミュー(Dromaius novaehollandiae)の内外寄生虫を調査した。アルパカの糞便材料ではEimeria 属とIsospora 属のコクシジウム・オーシストが確認された。前者は関東地方で行われた先行研究でも確認されていたが、後者は未確認であった。蠕虫卵としては線虫卵各種が検出され、これも先行研究で得られた知見と重複した。エミューのと殺個体の検査では蠕虫未検出であったが、糞便材料では線虫卵が検出されたので、新鮮な個体を用いた再検査が望まれた。道内で飼育される特用家禽のダチョウでは、ダチョウハジラミStruthioliperus struthionisの濃厚寄生が経験されるが、今回のエミューからは外部寄生虫が見出されなかった。

北海道日高地方におけるエゾシカ(Cervus nippon yesoensis)の内部寄生虫相および道内エゾシカ寄生日本産肝蛭(Fasciola sp.)の分布域について, 森 昇子; 三觜 慶; 鈴木 瑞穂; 萩原 克郎; 浅川 満彦, 北海道獣医師会雑誌, 58, (2) 8 - 11,   2014 02 , 北海道において、エゾシカ(Cervus nippon yesoensis)の個体数は増加の一途をたどっており、その生息域はヒトの生活環境および家畜の飼養領域にまで達している。このため、エゾシカの排泄物を介して家畜やヒトへも肝蛭などの寄生虫感染の波及が懸念される。本研究では、2011年11月から2012年9月において、北海道日高地方で得られたエゾシカの新鮮直腸便437検体について寄生虫検査を行い、内部寄生虫の感染状況を調査した。一般線虫卵の寄生率は95.0%、日本産肝蛭(Fasciola sp.)の寄生率は9.5%であった。今回の調査によりエゾシカにおける肝蛭寄生率は既報の釧路および十勝で報告されている寄生率と比較して低い値であった。この結果は、中間宿主であるヒメモノアラガイの道内分布域と関連性があると考えられた。

Parasitism of Solenopotes sp. cf. binipilosus on C. nippon was first established, although a record of Solenopotes sp. without any morphological data was presented in the other part of Hokkaido in the 1990's. Photographs of both insect species and their brief epidemiological comments are given.

As in February 2012, the total number of deaths and missing by this East Japan disaster was 19,139 and Ishinomaki was especially heavily affected; the number of death and missing 3,833 accounted for 20% of the total number in Japan. Rakuno Gakuen University sent 73 volunteers including 6 teaching and administrative staffs since April 2011 and its researchers started environmental risk assessment since July jointly with Ishinomaki City Council.
Stratified random sampling of the Chonai-Kai administrative units with two strata, the extremely damaged areas by Tsunami located within two kilometers from the sea shore and 11kms along coast were divided into eastern and western strata from Old Kitakami River, was performed and 20 out of 87 units were selected (the sampling fraction 24.4%). Sampling of sludge, water, flies, mosquitoes and rodents were conducted and the samples were served for microbiological, bio-chemical and chemical analyses.
Before Tsunami, livestock feed factories were located on the western seashore from Old Kitakami River and fish processing factories and freezer storages on the eastern. These feeds and fish were scattered on the Tsunami affected areas and these areas smelled very bad. Plenty of Musca and Spaerocerdae species flies that favor carbonates occurred on the western seashore and plenty of Phomiaregina flies that favor proteins on the eastern in June and July. City Council, volunteers and Self Defense Force removed fish and feeds and conducted spraying pesticides and eventually the smell and flies were controlled by August.
From the sludge and water samples, enterococcal bacteria and non-highly pathogenic Vibrio were isolated. Heavy metals were found but not in the alarming dose. Rodents seemed to die off other than one western area with canal.
From these results, risk was not so high but continuous monitoring is recommended. Sociological studies needs to be carried out to understand people’s intention of living in the areas again in order to present the overall risks.

Here, we evaluated a virus removal filter which has 15 nm pores. An antithrombin sample immediately prior to nano-filtration was spiked with prion material prepared in two different ways. The removal (log reduction factor) of prion infectivity using animal bioassays was >or=4.72 and 4.00 in two independent filtrations. However, infectivity was detected in both the pellet and supernatant following ultracentrifugation of the 15 nm filtered samples, indicating difficulty in complete removal. The data supports the conclusion that a certain amount of infectious prion protein is present as a smaller and/or soluble form (less than approximately 15 nm in diameter).

To examine infection kinetics, two litters of domestic pigs (A and B, each containing 10 piglets) infected naturally with HEV were studied until pigs were 6 months old. Maternal IgG and IgA antibodies were detected in litter A piglets, but not in litter B ones. All pigs shed HEV in feces when they were 30-110 days old, and 17 developed viremia at 40-100 days of age. Phylogenetic analysis revealed a highly close sequence of HEV genotype 3 in all pigs. The serum levels of specific IgG and IgA were similar in all pigs, although IgA was not detected in the feces. Interestingly, the onset of both viremia and seroconversion was delayed significantly in litter A pigs. The kinetics of fecal virus shedding was similar in both litters; shedding was not detected after the pigs were 120 days old. The differences in the infection kinetics between litters A and B suggested that maternal antibodies delayed the onset of viremia and seroconversion. Quantitative real-time reverse transcriptase-polymerase chain reaction revealed that HEV RNA in feces peaked 10 days after initial shedding of approximately 10(6.0) copies/g. The viral load was much lower in the serum than in the feces. At 200 days of age, HEV RNA was found in the internal organs of 3 out of 13 pigs. These study findings improve the understanding of the dynamics of natural HEV transmission in pigs, which could help in controlling virus transmission from pigs to humans.

Serum samples from raccoons were examined using ELISA and Western blot assays to detect the presence of serum antibodies that react specifically to BDV antigens. Among 549 investigated individuals, eleven (2.0%) showed a positive reaction to BDV antigens. Brain tissue samples from five individuals were subjected to RT-PCR, which detected BDV sequences in three of them. Sequence analysis revealed a high degree of genetic conservation between BDV sequences derived from raccoons and previously published sequences derived from other animal species.

The data showed that urinary 8-OHdG ranged from 4.3 to 193.1ng/mg creatinine. An individual range of median value was 6.8-52.4ng/mg creatinine. No statistically significant effect was found for infection of protozoan parasites (Balantidium coli and Isospora sp.) without any symptoms (>0.05).

We have previously reported that bovine colostrum contains high levels of IFN-gamma as well as immunoglobulin. Lymphocytes are potent producers of IFN-gamma, so establishing the lymphocyte population in colostrum will help to identify the source of colostral cytokines. In this study, we used flow cytometric analysis to quantify the population of three types of lymphocytes found in colostrum; namely, CD4 (Th) cells, CD8 (cytotoxic T) cells, and gammadelta-T cells. We also quantified the concentration of colostral IFN-gamma using ELISA. IFN-gamma concentrations were measured in colostrum obtained from 96 healthy Holstein cows; the levels tended to decrease on the first day post-parturition. Flow cytometric analysis showed that many gammadelta-T- and CD8-positive cells were present in the colostrum, and that the CD4/CD8 ratio was low. The ratios of CD8- and gammadelta-T-positive cells to cells of other types decreased during the 5 days after parturition, but that of CD4-positive cells showed no change during the observation period. To identify IFN-gamma-expressing colostral lymphocytes, we used magnetic separation technology to separate the lymphocytes (CD4, CD8 and gammadelta-T) from colostral cells, then examined them for IFN-gamma mRNA expression with real-time PCR (RT-PCR). RT-PCR analysis revealed potent expression of the IFN-gamma gene in CD8-positive cells, reaching higher levels than in CD4- or gammadelta-T-positive cells. These results suggest that the CD8-positive T cells in colostrum play a role as producers of IFN-gamma.

In this study, we examined the anti-HEV antibodies and HEV-RNA in sera from 173 of female sika deer in the park. The reactivity to HEV antigen in the serum samples were low levels. The detection of HEV-RNA in sera from the deer revealed no positive samples by RT-PCR analysis. In conclusion, we could not able to identify the HEV antibody and HEV-RNA positive individual in the sika deer which inhabited Nara Park. Therefore, the possibility of the HEV infection in the deer of the park would be extremely low.

Serum samples from macaques were examined by the ELISA, Western blot and immunofluorescence assays to detect the presence of serum antibodies that react specifically to BDV antigens. Among 49 investigated individuals, 6 (12.2%) showed positive reaction to BDV antigens. RT-PCR studies detected BDV sequences in brain tissue of one case among four seropositive cases examined. Sequence analysis revealed a high degree of genetic conservation between BDV sequences derived from Japanese macaques and those documented for other animal species. Nevertheless, phylogenetic analysis revealed unique differences between macaque and other species derived BDV sequences.

In this study, we examined the distribution and multiplication of HEV genotype 3 in two piglets at the early phase of intravenous infection, and also examined the virus distribution in a naturally infected pig. We developed real-time RT-PCR to determine copy numbers of the HEV genome in the sera, feces and organs. HEV-RNA was detected in serum from a pig transiently for 7 days post-inoculation (dpi). The HEV copy numbers in the feces appearing 7 dpi were increased to 3.4x106 copies/g at 14 dpi. In contrast, the higher copy numbers of HEV were widely distributed in the tissue organs from naturally HEV-infected pig. The study showed that intravenously inoculated HEV was distributed in several restricted tissues, such as in the liver and intestine.

IL1R1 contributes to IL-1 signaling, whereas the IL1R2 has no signaling property and acts as a decoy for IL-1. In this study, we developed a bovine IL1R2-specific sandwich ELISA to examine the sIL1R2 concentration in serum and milk from dairy cows. The concentration of colostral IL-1beta was examined to estimate the correlation to sIL1R2. The results showed that the sIL1R2 concentration in sera and milk changes with the stages of lactation. The serum sIL1R2 concentrations were 5.56+/-0.69 ng/ml (colostrum), 3.14+/-0.72 ng/ml (the early stage of lactation) and 5.76+/-1.25 ng/ml (the late stage of lactation). The milk sIL1R2 concentrations were 1.83+/-0.47 ng/ml (colostrum), 0.73+/-0.22 ng/ml (the early stage of lactation) and 2.92+/-0.56 ng/ml (the late stage of lactation). The concentrations of IL1R2 in sera and milk were significantly higher at the late stage of lactation and colostrum than that of the early stage of lactation. The reduction rates of sIL1R2 levels from the colostrum to the early stage of lactation were 43.6% (serum) and 61% (whey). IL-1beta was detected in all the colostrum (995.9+/-346.6 ng/ml). Significant correlation was observed between the levels of colostral IL-1beta and IL1R2 (r=0.75).

Natural science, 29, (2) 171 - 175,   2005 04 , 初乳は新生子の免疫機能を活性化するサイトカインや経口寛容成立に重要と考えられるサイトカインを含んでいるため、新生子の感染症感受性を大きく支配する要因であると考えられる。そこで、初乳摂取12-24時間の新生子血清中のIL-1β、IL-1ra、IL-6、IFN-γ、GF-β1およびNF-αをELISAで測定し、生後4ヶ月間の疾病発生状況との関連を調べた。ホルスタイン種ではIL-1β、IL-1ra、NF-α濃度が健康群より罹患群で有意に低いことが観察された。黒毛和種では同様の傾向を認めたが有意差はなかった。この成績は初乳摂取後の新生子血清サイトカインレベルは疾病対策上有効な指標となりうることを示唆するものであると考えられた。

In the present study, we quantified the serum IFN-gamma in splenectomized (SP) and non-splenectomized (NSP) cows infected with Theirelia sergenti (TS) to investigate whether the spleen plays a role in the Th1-type cytokine responses in cows following parasite infection. A transient increase in IFN-gamma was observed in the early stages of infection in the NSP cowl., and the cows did not develop parasitemia. In contrast, the SP cows showed no IFN-gamma response at the early stage of infection. and the cows developed parasitemia following anemia. The NSP cows produced IgG1 and IgG2 antibodies to TS-specific. whereas the SP cows showed increases only in IgG1. Increased PBMC proliferation by cocanavalin A stimulation was observed concomitant with the IFN-gamma response. Pretreatment with IFN-gamma suppressed the propagation of TS in the SP cattle. These results suggest that the spleen plays an important role in the resistance to TS infection. leading to a Th1-type immune response. IFN-gamma might contribute to the activation of immunocytes and play an important role in the immune response to resist TS proliferation. (C) 2004 Elsevier B.V. All rights reserved.

It is important to predict postpartum reproductive diseases early and to develop prophylaxis. The objectives of this study were to demonstrate changes in the peripheral blood concentration of interleukin-6 (IL-6) before parturition, which was mainly produced by T helper 2 type (Th2) cells, and to investigate a correlation between the IL-6 concentration and the occurrence of the postpartum retained placenta, endometritis and/or follicular cyst in dairy cattle. Twenty-seven Holstein-Friesian cows were used for this study. Thirteen had no clinical disease, 8 had retained placenta, 4 were diagnosed with endometritis by vaginal inspection, and 2 were diagnosed with follicular cyst by rectal palpation at I and 2 months after parturition. Blood samples were collected 60 days pre- and post-partum. They used for IL-6, progesterone (P-4) and estradiol-17beta (E-2) concentration determination. This study showed that the IL-6 concentration prepartum was higher than postpartum. Low levels of IL-6 and P-4 in peripheral blood prepartum tended to affect retained placenta and a high level of IL-6 prepartum tended to affect endometritis. These results indicate that measurement of change in the IL-6 concentration during pregnancy is one useful tool for predicting crisis in postpartum reproductive diseases in dairy cattle.

To verify the possibility, this study was conducted mainly on parasites that included protozoan, helminth and arthropod, and other pathogens including Rickettsia and the Borna virus of 50 free ranging Japanese macaques in the peninsula. Consequently, 5 arthropod and nematode parasite species common to the Japanese macaque, namely Pedicinus obtusus, P.eurygaster, Streptopharagus pigmentatus, Strongyloides fulleborni, and Trichuris trichiura, were obtained in this survey. It is the first record of sites where Pedicinus spp. are seen in a host species.Coccidia and Babesia sp. were not obtained. Coxciella and the Borna virus from the same host samples will be published elsewhere.

To determine whether these new types of parasites are distributed in places surrounding Japan, an epizootiologic survey was undertaken in three geographically distant areas in northeastern Eurasia; South Korea, Vladivostok in Russia, and Xinjiang in China. Blood samples were collected from a total of 387 animals comprising 24 species. DNAs extracted from the samples were tested by nested PCR targeting babesial nuclear small-subunit rRNA gene (rDNA), which revealed that small rodents harboring B. microti exist in all three survey areas. Sequence analysis showed that all PCR-positive samples had rDNA sequences virtually identical to that of U.S.-type B. micron. However, when beta-tubulin gene sequences were compared, evident geographic variations were seen. By use of primers specific for each of the beta-tubulin genes of Kobe-, Hobetsu-, and U.S.-type parasites, a type-specific PCR was developed. Parasite with Hobetsu- or Kobe-type sequence was not detected from any of the three survey areas. These findings suggest that U.S.-type B. microti is widely distributed among small wild mammals in temperate zones of not only North America, but also Eurasia, whereas that Hobetsu- and Kobe-type parasites may be uniquely distributed in Japan.

IL-1beta concentrations in the serum of the calf with BLAD at age 143-162 days ranged from 8.7 to 16.6 ng/ml, whereas the values were less than 2.7 ng/ml in control calves. Serum IL-6 (0.04 ng/ml) was only detected on the 1st day when the animal was diagnosed with the BLAD. IL-1beta and IL-8 mRNA expression on neutrophils from the affected calf appeared to be similar to those of controls. Serum cytokine levels and their mRNA expression on neutrophils from the calf with BLAD appeared to be little affected by the deficient expression of beta(2)-integrin on leukocytes, and are considered to be modulated by the inflammatory stimuli.

Analysis of serum cytokine profile in a holstein heifer with leukocyte adhesion deficiency which survived for long period, H Nagahata; K Hagiwara; H Higuchi; R Kirisawa; H Iwai, JOURNAL OF VETERINARY MEDICAL SCIENCE, 64, (8) 683 - 687,   2002 08 , 10.1292/jvms.64.683, Serum cytokine levels and their expression of mRNA on neutrophils from a bone marrow (BM) transplanted heifer with leukocyte adhesion deficiency were evaluated. The clinical condition of the affected heifer was relatively stable after BM-transplantation. Persistent hyper gamma-globulinemia and increased serum immunoglobulin G (IgG) concentrations were monitored longitudinally. The concentration of interleukin (IL)-1beta in serum from the affected heifer ranged from 15.8 to 321.7 ng/ml, and maximum concentration occurred at the time which coincided with peak IL-6. Serum levels of IL-6 ranged from 0.32 to 27.9 ng/ml, and they appeared to be associated with the increment of serum IgG in the affected heifer. mRNAs for IL-10, IL-6, IL-8 and granulocyte and macrophage colony stimulating factor (GM-CSF) were increased in neutrophils from the affected heifer compared to controls. Persistent hyper gamma-globulinemia of the affected heifer appeared to be associated with enhanced mRNA expression for IL-6 and its serum levels. These findings suggest that humoral immunity of the affected heifer is activated and the production of neutrophils appears to be enhanced under the incapability of beta(2) integrin-mediated functions of phagocytic cells.

The serological study disclosed the presence of antibodies only to BDV-p40 or -p24 in 19.2% (24/125) and 3.2% (4/125) of culled horses, respectively. Antibodies to both BDV-p40 and -p24 were found in 24.0% (30/125) of these horses. Of particular note was the finding that locomotorium disorders were detectable at a significantly higher rate in BDV antibody positive horses than that in the seronegative horses. These results imply that BDV infection may possibly contribute to an increase in the incidence rate of locomotorium disorders in race horses. © 2002 Elsevier Science B.V. All rights reserved.
Interferon-γ and Tumor Necrosis Factor-α Levels in Sera and Whey of Cattle with Naturally Occurring Coliform Mastitis, Keiichi Hisaeda; Katsuro Hagiwara; Junko Eguchi; Hitoki Yamanaka; Rikio Kirisawa; Hiroshi Iwai, Journal of Veterinary Medical Science, 63, (9) 1009 - 1011,   2001 09 , 10.1292/jvms.63.1009, Concentrations of interferon-γ (IFN-γ) and tumor necrosis factor-α (TNF-α) were determined in serum and whey samples from cattle with naturally occurring coliform mastitis for two weeks after onset using bovine INF-γ- and TNF-α-specific ELISA. In serum and whey samples from healthy cows, IFN-γ was almost undetectable and TNF-α was detected at low levels. At the onset of illness, INF-γ in sera and whey and TNF-α in whey from the mastitic cows were significantly higher than their respective values in healthy cows. Concentrations of IFN-γ and TNF-α in whey from mastitic cattle decreased significantly as the cows recovered.

A total of 124 individuals belonging to 24 species were examined, and an acanthocephala and/or nematodes were obtained from the following 15 host species, i.e., Dipus sagitta [Ds] [Abbreviation of host name], Euchoreutes naso [En], Allactaga sibirica [Asi], Meriones meridianus [Mme], M. tamariscinus [Mt], Rhombomys opimus [Ro], Cricetulus migratorius [Cm], C. eversmanni [Cev], Microtus oeconomus [Mo], Lagurus lagurus [Lla], L. luteus [Llu], Alticola argentatus [Aa], Clethrionomys rutilus [Cr], Microtus arvalis [Ma], and Apodemus uralensis [Au]. To date, 1 acacthocephalan species, Meniliformis sp. [Cev] [abbreviation of infected animals], and at least 19 nematode species, i.e., Eucoleus sp. (gastrica?) [Au], Eucoleus sp. [Asi], Aonchotheca murissylvatici [Cr], Calodium hepaticum [Au], Trichuris muris [Llu], Trichuris sp. (rhombomidis?) [Mt], Dentostonella translucida [Mme), Aspiculuris tetraptera [Ds], Syphacia petrusewiczi [Cr], Syphacia sp, (or spp.) [Mt, Cm, Cev, Lla and Ma], Rhabditis (Pelodera) orbitalis (3rd larvae) [Lla, Cr, Ma and Mo], Heligmosomoides yorkei [Cm], H. polygyrus [Au], Heligmosomum (Paraheligmosomum) mixtum [Cr and Ma], Heligmosomum (Heligmosomum) sp. [Aa and Mo], Subulura citelli [En], Pterygodermatites sp. [Cr], Litomosa vite [Ro], and spirurid unknown species [Cr] were obtained. Although most species have been reported from the former USSR or outer Mongolia, several species are first records in Xinjiang. Photographs of these nematodes are presented and helminthological studies of the parasitic nematodes obtained from the wild small mammals in Xinjiang are briefly reviewed.

Here, authors examined oral transmission of colostral IL-1 beta and its immunological effects on the neonatal calves. Biotin-labeled recombinant bovine (rb) IL-1 beta was administered orally to newborn calves and monitored in the serum, The results disclosed the passive transfer of colostral cytokines via the oral route, and a potent increase in white blood cell (WBC) count was observed in all calves administered with rbIL-1 beta. Oral administration of IL-1 beta significantly increased the proliferation of peripheral blood mononuclear cells (PBMCs) stimulated with concanavalin A, and the O-2(-) production of stimulates neutrophils in newborn calves, These results suggest that the oral administration of IL-1 beta has an immunostimulatory activity in the newborn calf. In the present study, we investigated the effect of IL-1ra on the mitogenic response of calf peripheral blood mononuclear cells (PBMC) stimulated by concanavalin A (ConA), which was mediated by IL-1. Pretreatment of PBMC with recombinant bovine (rb) IL-1ra alone significantly suppressed the proliferation of ConA-stimulated cells. However, in the presence of rbIL-1 beta, the suppressive activity of rbIL-1ra was counteracted. These results suggest that coexistence of IL-1ra with IL-1 in colostrum may have no effect on the activation of the neonatal immune system by IL-1 beta.

The serological study disclosed the presence of antibodies to both BDV-p24 and -p40 in the horses (20%) and sheep (25%), whereas no apparent positive reaction was detected either in cattle or rodents. In this study, we quantified the IL-6 concentrations in the serum and milk whey from 94 dairy cows with acute clinical mastitis and 55 healthy lactating cows. The IL-6 concentrations in serum from mastitic cows were significantly higher on the first day of illness compared to those of normal cows. Higher concentrations of IL-6 were also detected in the whey from mastitic cows, whereas low concentrations of IL-6 were detected in both serum and whey samples from normal cows. IL-6 concentrations in the serum taken at the onset of illness from cows that later required euthanasia were significantly higher than those in samples from cows that later recovered. These results suggest that serum IL-6 concentrations may be of prognostic value in identifying cows with severe mastitis.

To confirm that colostrum contains a variety of cytokines with immunomodulatory properties, we established a bovine cytokine specific ELISA and five cytokines (IL-1 beta, IL-6, TNF-alpha, INF-gamma or IL-1 receptor antagonist, IL-1ra) in the whey samples from cows at different stages of lactation were monitored. The expression of cytokine mRNAs (IL-1 beta, IL-6, TNF-alpha and INF-gamma) in the colostral cells was detected by RT-PCR. The concentrations of cytokines in colostrum were significantly higher concentrations than those in the mature milk. A positive correlation was observed between the concentrations of IL-1ra and IL-1 beta in the colostrum samples. In conclusion, colostrum contains high levels of cytokines that could be produced and secreted in the mammary gland and that may have an immunomodulatory activity and influence neonatal immunity. The BDV nucleotide sequences were identical in the mare and fetus in the second open reading frame (ORF). This is the first report of the possible vertical transmission of BDV in a horse. The horses inoculated with this V26 virus were shown to raise neutralizing antibodies against V70 without any viremia. Here, we investigated the in vitro and in vivo replication ability of V26. Comparison of the long-terminal repeat (LTR) sequences between V26 and V70 revealed a large insertion within the LTR U3 hypervariable region of V26. V26 with the mutation in the LTR showed much higher promoter activity in vitro than V70. This is consistent with the much higher replication rate of V26 in horse primary macrophage cultures compared with V70. In sharp contrast, we failed to identify the V26-specific LTR sequence by PCR, at least in sequential samples of plasma or peripheral blood mononuclear calls derived from three horses until day 62 after V26 inoculation. In contrast, antibody responses to EIAV were observed in all horses. The results suggest that the replication ability of V26 in vivo is extremely low. When one of the horses was subsequently challenged with cell-associated V70, it was found that the horse became PCR positive for EIAV. There was no LTR mutation in EIAV genome in samples periodically prepared from the V70-challenged horse. Thus it was suggested that the LTR mutation in EIAV, which occurs during serial passage in vitro, affects EIAV replication in vitro and in vivo. (C) 2000 Academic Press.

A (1 --> 4)-linked homopolysaccharide composed of N-acetyl, 6-O-carboxymethyl glucosamine (CM-chitin) showed no inhibitory activity but a 3,6-O-sulfated derivative of CM chitin (SCM-chitin III) showed potent hemagglutination inhibition activity. A N-sulfated derivative of N-deacetylated CM chitin (SCM-chitosan) however, was not effective. Chondroitin sulfate and dermatan sulfate, both of which consisted of disaccharide repeats of (1 --> 3)-linked uronic acid and galactosamine sugar residues (--> 4GlcAb1/IdoAal --> 3GalNAcb1), did not interfere with the red blood cell (RBC) binding by T. sergenti merozoite. This virus induced encephalitis in experimentally infected adult rats, but in newborn rats BDV established a persistent, tolerant infection with no apparent clinical signs. Here, we report evidence that newborn Mongolian gerbils (Meriones umguiculatus) are more susceptible to experimental intracranial inoculation of horse-derived BDV in persistently infected MDCK cells, compared with similar inoculation in newborn rats. All inoculated newborn gerbils, but not rats, died 30 days after infection. Reverse transcriptase-polymerase chain reaction amplified BDV-specific sequences in several regions including the brain. Histopathological analysis revealed apparent inflammatory reactions in the brains of inoculated gerbils but not rats, although similar levels of BDV RNA were detected in both gerbil and rat brains. BDV-specific antigen and RNA were identified predominantly in neurons in the brains by immunohistochemistry with antibodies to BDV and in situ hybridization with BDV-specific riboprobes, respectively BDV in the gerbil brain was easily rescued by co-cultivation of the brain homogenate with human oligodendroglioma cells. Thus, gerbils seem to be a useful animal model for studying BDV-induced pathogenesis in the brain. (C) 1999 Elsevier Science Ltd. All rights reserved.

The deduced amino acid sequence of bovine IL-1ra demonstrated 80%, 78%, 78%, 77% and 76% homology with human, mouse, rat, rabbit and equine sequences, respectively. Recombinant bovine IL-1ra produced in Escherichia coli suppressed the growth inhibitory activity of bovine IL-1 beta on A375 cells in a dose-dependent manner, indicating that the present bovine IL-1ra cDNA encodes biologically active proteins. A mouse hybridoma cell producing antibody against Holstein RBC was established for distinguishing Holstein RBC from Japanese Black RBC by a immune-staining method. T. sergenti piroplasms in RBC were stained by 4',6-diamidino-2-phenylindole. With the aid of these techniques the piroplasm transmission from one to another race of Bo-RBCs could successfully be quantified in SCID mice.

In this study we demonstrated BDV RNA signals by polymerase chain reaction only in restricted regions of the brain from horses with locomotor disease. Four of six horses examined showed apparently positive reactions for anti-BDV antibodies. Specific regions of the brain of these four horses were positive for BDV RNA but the internal organs, lymph nodes, and PBMCs were negative. Histological studies of their brains revealed no apparent histological abnormalities such as inflammatory reactions. These results suggest that BDV chronically infects certain restricted regions of brain in seropositive horses.

The BDV prevalences by immunoblotting and/or reverse transcriptase PCR were 0% (0 of 19) in newborns (<1 month old), 51.7% (15 of 29) in lambs (1 to 6 months old), and 36.7% (11 of 30) in adults (>2 years old), Among animals positive for BDV, 60% of lambs and 45.5% of adults contained BDV RNA in PBMCs while 46.7% of lambs and 90.9% of adults contained specific antibodies to BDV, Thus, it is suggested that virus replication in the blood, as observed in lambs, is usually reduced in adulthood by raising immune responses to BDV.

Here me have focused on the effect of heparin, which has an inhibitory effect on RBC invasion by Plasmodium falciparum, and demonstrated for the first time that bovine RBC invasion by T. sergenti was inhibited by heparin, Further, analysis of this mechanism showed that bovine RBC agglutination, by purified T. sergenti merozoites, mas inhibited by heparin and low molecular weight (LMW) heparin, Moreover, hemagglutination was inhibited by treatment of the merozoites with heparinase, These results suggest that merozoites have heparin-like molecules on their surface which may be one of the important factors for attachment to RBCs. The seroprevalence determined using recombinant BDV p24 protein was 20.3%. The prevalence of BDV RNA in peripheral blood mononuclear cells (PBMCs) determined by polymerase chain reaction for p24 was 10.8%. Cell fraction of the PBMCs revealed the presence of BDV RNA in the T cell-enriched fraction. Thus, BDV infection is wide-spread even among healthy cattle.

Free merozoites (1 x 10(8)) were incubated with erythrocytes (2 x 10(7)) in RPMI-1640 containing various concentrations of fetal bovine serum (FBS). The invasion assay was examined at various temperatures and pH concentrations. The results showed that merozoite invasion was accelerated by the addition of FBS and by incubation at 37 degrees C at pH 6.5-7.5. To investigate how enzymatic activity contributes to the entry of T. sergenti merozoites, the inhibitory effect on the merozoite invasion was examined by using protease inhibitors. An inhibitory effect on the invasion of merozoites was observed in the medium containing 1,10-phenanthroline and ethylenediaminetetraacetic acid disodium salt which are classified as metalloprotease inhibitors. The results suggest that FBS plays an important role in the effective invasion of merozoite into erythrocytes and that enzymatic activity may affect the invasion of merozoite into the host cell.

SERUM FROM THEILERIA SERGENTI-INFECTED CATTLE ACCELERATES THE CLEARANCE OF BOVINE ERYTHROCYTES IN SCID MICE, K HAGIWARA; M TSUJI; C ISHIHARA; M TAJIMA; T KUROSAWA; K TAKAHASHI, PARASITOLOGY RESEARCH, 81, (6) 470 - 474,   1995 07 , Anemia is a major clinical sign of Japanese bovine theileriosis caused by Theileria sergenti. To investigate the possible factors causing anemia in cattle, we developed a clearance test for bovine erythrocytes (Bo-RBC) in severe combined immunodeficient (SCID) mice. Clearance of Bo-RBC in the SCID mice was significantly accelerated when the mice were inoculated with a serum sample obtained from an infected calf during a highly parasitized phase but not when they were injected with a serum sample obtained during the convalescence phase. Acceleration of the clearance of Bo-RBC was also observed in mice treated with merozoite extract. Furthermore, the clearance of Bo-RBC that had been treated with merozoite extract was accelerated. A significant hemolytic activity in infected serum (highly parasitized phase) was observed. Activities sufficient to accelerate the clearance of Bo-RBC in SCID mice and to induce in vitro hemolysis of Bo-RBC were also observed with a merozoite extract from T. sergernti. The results suggest a possible linkage between the in vitro hemolysis of Bo-RBC and the acceleration of Bo-RBC clearance in SCID mice.
TRANSFUSION WITH XENOGENEIC ERYTHROCYTES INTO SCID MICE AND THEIR CLEARANCE FROM THE CIRCULATION, C ISHIHARA; M TSUJI; K HAGIWARA; K HIOKI; J ARIKAWA; AZUMA, I, JOURNAL OF VETERINARY MEDICAL SCIENCE, 56, (6) 1149 - 1154,   1994 12 , 10.1292/jvms.56.1149, We have previously demonstrated that the red blood cells (RBCs) in the blood circulation of SCID mice could be almost completely substituted with bovine RBCs by means of repeated transfusions. In the present study, transfusion experiments were carried out with RBCs from various animal species to investigate the ability of SCID mice to accept xenogeneic RBCs. Bovine (Bo), equine (Eq), human (Hu) and murine (Mu) RBCs were labeled with a fluorescent dye to trace their trafficking in the blood stream. Following the intravenous injection these RBCs were cleared from the circulation at various rates; the 1/100 reduction time in the labeled cell counts was 2 to 7 days, 4 to 7 hr and 1 to 2 hr with Bo-, Hu- and Eq-RBCs, respectively. In contrast, labeled Mu-RBCs from not only syngeneic but also allogeneic mouse strain were able to stay over 50 days. The difference in clearance rates was attributable to the difference in uptake of the RBCs by the splenic macrophages. The clearance rates of labeled RBCs were significantly decreased by co-transfusion with unlabeled RBCs of the same species, indicating that xenogeneic RBCs were recognized by the reticulo-endothelial system of SCID mice in a species-specific fashion. Furthermore, at least in the case of Hu-RBC, the complement component 3 may play a role in the Hu-RBC clearance in SCID mice since C3 deposition was observed on Hu-RBCs but not on Bo- and Eq-RBCs.
THEILERIA-SERGENTI INFECTION IN THE BO-RBC-SCID MOUSE MODEL, K HAGIWARA; M TSUJI; C ISHIHARA; M TAJIMA; T KUROSAWA; H IWAI; K TAKAHASHI, PARASITOLOGY RESEARCH, 79, (6) 466 - 470,   1993 08 , We have previously developed the Bo-RBC-SCID mouse model for Theileria sergenti infection. In the present study, this model was further examined to delineate the mode of parasite infection. The Bo-RBC-SCID mice were prepared by periodically transfusing uninfected bovine erythrocytes (Bo-RBCs) into splenectomized SCID mice via the intraperitoneal (i.p.) route. The mice, separated into three groups, were inoculated i.p., intravenously (i.v.), or subcutaneously (s.c.) with T. sergenti-infected Bo-RBCs. Examination of samples of peripheral blood demonstrated that the parasite infected mice inoculated via any one of the three routes. The mice inoculated i.v., however, developed parasitemia earlier than those inoculated i.p. or s.c. When Bo-RBC-SCID mice prepared without splenectomy were infected with T. sergenti, a high-level parasitemia appeared only once. After that, not only the level of parasitemia but also the number of Bo-RBCs in the peripheral blood rapidly decreased despite the continuation of Bo-RBC transfusions. The results suggest that T. sergenti proliferates primarily in the circulating blood in Bo-RBC-SCID mice and that in response to the parasites growth, the spleen may play an important role in the removal of both parasitized and unparasitized Bo-RBCs from the blood circulation.
THE BO-RBC-SCID MOUSE MODEL FOR EVALUATING THE EFFICACY OF ANTI-THEILERIAL DRUGS, K HAGIWARA; M TSUJI; C ISHIHARA; M TAJIMA; T KUROSAWA; H IWAI; K TAKAHASHI, INTERNATIONAL JOURNAL FOR PARASITOLOGY, 23, (1) 13 - 16,   1993 02 , We have previously developed a mouse model which allowed the proliferation of Theileria sergenti in severe combined immunodeficiency (SCID) mice with circulating bovine erythrocytes (Bo-RBC). In the present study, this model was utilized to test the efficacy of anti-theilerial drugs. Bo-RBC-SCID mice were created by giving periodic transfusions of T. sergenti-free Bo-RBC, and subsequently infecting with T. sergenti. Three anti-protozoal compounds, Pamaquine (Yamanouchi Pharmaceutical Co. Ltd), Ganaseg (Japan CIBA-GEIGY Ltd) and Buparvaquone (Coopers Animal Health Ltd), were subcutaneously administered into the mice at doses recommended for cattle therapy. Blood examinations demonstrated that all three drugs significantly reduced the level of parasitemia although Ganaseg was effective only at a dose five times higher than that recommended for cattle therapy. Administration of the drugs neither caused any sign of acute toxicity nor changed the rate of Bo-RBC in the SCID mice's circulating blood cells. The results indicate that the Bo-RBC-SCID mouse model may offer a useful in vivo system for evaluating the efficacy of anti-protozoal drugs against T. sergenti.
THE DETECTION OF A MUTATION OF CD18 GENE IN BOVINE LEUKOCYTE ADHESION DEFICIENCY (BLAD), M TAJIMA; M IRIE; R KIRISAWA; K HAGIWARA; T KUROSAWA; K TAKAHASHI, JOURNAL OF VETERINARY MEDICAL SCIENCE, 55, (1) 145 - 146,   1993 02 , 10.1292/jvms.55.145, Two calves (5 and 9 months old) affected with pneumonia and gingivitis were also diagnosed as having bovine leukocyte adhesion deficiency (BLAD). The gene of leukocyte adhesion molecule CD18 in these BLAD calves and their dams (carrier) were examined by means of polymerase chain reaction (PCR) and digestion of restriction endonuclease. The splicing in mRNA coded CD 18 reported in human LAD was not recognized in BLAD on the basis of the results of PCR amplification. The region including the portion of point mutation, which corresponded to the region reported in the human patient, was amplified by PCR, and the PCR product was then digested with TaqI. An obvious difference was recognized in the pattern of digestion among healthy calves, BLAD calves and their dams. In BLAD, therefore, the point mutation reported in human patients was strongly suggested. Moreover it may be a method able to be used in detecting the carrier.
THEILERIA-SERGENTI PROLIFERATES IN SCID MICE WITH BOVINE ERYTHROCYTE TRANSFUSION, M TSUJI; K HAGIWARA; K TAKAHASHI; C ISHIHARA; AZUMA, I; WA SIDDIQUI, JOURNAL OF PARASITOLOGY, 78, (4) 750 - 752,   1992 08 , The unavailability of in vitro or in vivo experimental systems has been the major factor hampering the progress of research studies on Theileria sergenti, causative agent of theileriosis, a major disease of cattle in Japan. We report the first successful propagation of T. sergenti in SCID mice into which uninfected bovine erythrocytes (Bo-RBC) were supplied periodically. The infectivity of T. sergenti proliferated in an SCID mouse was ascertained by successful transfer of infection into another SCID mouse into which uninfected Bo-RBC were supplied periodically.

日本学術振興会, 科学研究費助成事業 国際共同研究加速基金(国際共同研究強化(B)), 新興感染症パンデミック予防に向けたタイ国における未知病原体の潜在的蔓延状況調査, 20201027, 20240331, 大道寺 智; 西岡 敬介; 村越 ふみ; 萩原 克郎, 本研究の目的は、タイ山間部において蔓延している病原体をヒト、野生動物、媒介節足動物から網羅的に同定することで、今後流行を起こす可能性がある未知病原体を検出することである。国際研究グループ(タイ)との連携により実施する。新型コロナウイルスの影響で予定していた渡航ができなくなったため国内での活動を余儀なくされた。そのため本年度はタイの病原体の検出に先立ち、前年度までに改良を続けてきた病原体検出技術を用いて、国内で採取した節足動物を用いた解析を行うこととした。 京都府下で採取した野外マダニより核酸抽出し、16S rRNA遺伝子をPCR にて増幅し、次世代シーケンサーにて、マダニの保有する細菌叢について解析を行った。その結果、今回捕獲した野外マダニの細菌叢として、腸内細菌科細菌の他、リケッチア属、コクシエラ属、ミクロコッカス属、エーリキア属、スフィンゴモナス属などの細菌が検出された。中でもリケッチア属、コクシエラ属といった病原細菌も比較的高率に検出され、更なる解析の必要性が示唆された。一方で、同様に京都府下で採取した野外蚊より核酸抽出し、これまでに改良してきたフラビウイルス属をターゲットとしたユニバーサルプライマーを用いたPCR法にてフラビウイルス遺伝子を複数検出した。現在、塩基配列解析を含めさらなる解析を進めている一方で、ウイルス分離を試みている。 また上記のような実際の実験を推し進める一方で、共同研究者どうしでリモート会議を行い(国内)、今後の研究の進め方などについて協議を行った。タイ側共同研究者とも同様にリモート会議を行った。

Thus, persistent infection of the hypothalamus with BoDV-1 reduces GnRH expression and regulates the secretion of the normal gonadotropic hormones FSH and LH from the pituitary gland, resulting in abnormal reproductive organ development and prolonged sexual cycle. The abnormal sexual cycle due to decreased GnRH expression observed in the rat model suggests that the BoDV-1 persistently infected cows consist of few progesterone-producing cells in the corpus luteum and many non-secreting cells, confirming that the physiological GnRH stimulation response is not sufficient.

By analyzing in the mechanism of phage resistance in bacteria, this study clarified the mechanism of phage resistance and laid the foundation for designing strategic phage cocktail therapy. In addition, for the first time in Japan, a successful clinical trial of phage therapy was conducted on clinical cases in dogs and cats. We believe that the details of the phage resistance of bacteria in this clinical trial and the design of phage cocktails to deal with it will be valuable knowledge for future clinical applications in humans, and we plan to report them widely with the publication of a paper.

We have targeted the soot as a model air pollutant, which often generated from incomplete combustion of various fuels, and ozone as environmental stress factor to measure the effect on the viability of bacteria in a simulation reaction chamber. From the result, the soot and bacteria co-existing condition showed less influence of ozone stress. Furthermore, we have demonstrated that examination of soot and/or other organic compounds to contribute to maintaining the viability of airborne bacteria.

The results indicated that in-uterus persistently infected fetus moved as pregnant cow with the dam among the herd, then the new prevalence was confirmed. It seemed to be important for the prevention to keep the BVDV-free status besides vaccination.

The asian dust which occurred in the Mongolian Gobi desert arrived in Hokkaido area through the Westerly Wind without spreading in the meridian direction, according to meteorological model. We were also able to confirm occurrence / transport of Asian dust from LIDAR (Light Detection and Ranging, Laser Imaging Detection and Ranging) and artificial satellites CALIPSO. From aerosol samples acquired in Mongolian Gobi desert and Sapporo, they were biologically identified between them.

There is a demand for new strategies to treat bacterial infections. In this research, we investigated the ‘phage therapy for bacterial infection’ and ‘phage vaccine’. We examined the use of phage to treat horse bacterial keratitis as a research of phage therapy. We isolated phages toward P. aeruginosa causing horse keratitis. The phages in this study could completely prevent the keratitis caused by P. aeruginosa in a keratitis mouse model. In the phage vaccine, we constructed phage with surface antigen of Mycoplasma.

Further understanding of interaction between SPM and bioaeosol in the atmosphere may contribute to prevent spread of pathogenic microbes through air. This work investigates the effect of marine deposits and some other type of SPM to attribute a survival of biological aerosol in the tropospheric condition. Experiments were carried out with a reaction chamber to examine different types of SPM and bioaerosol in a laboratory. Results indicated that the SPM of marine deposit and model enteric bacteria had significantly higher survival rate than other SPMs examined. The different SPMs may play a significant role for the survival of airborne bacteria and other pathogenic microbes. It is necessary to further investigate the interactions between SPM and bioaerosols to understand factor influencing spread of pathogens to safeguard public health.

As a result, a HEV-RNA was detected in approximately 47% of individual livers out of 629 Yezo deer (Cervus nippon yesoensis) which we investigated and showed a viral gene in 12% more individual feces. Also, after investigating the palm civet (Paguma larvata) which inhabited Honshu, we detected the viral genome of 30% in feces, 53% in liver. It was suggested that a HEV habitually resided to these wild animals. Also, the deer HEV was transmitted by the experimental infection to a pig, and HEV was isolated from the feces of some individuals and confirmed infectious viral excretion from the infected deer.

Better basic knowledge of these highly pathogenic viruses and novel technologies such as reverse genetics will help us to generate effective prophylactic and/or therapeutic strategies to control the disease load. We have developed the reverse genetics of negative strand RNA viruses and were analyzing the mechanisms behind the replication and cytopathicity of these viruses. Primary airway epithelial cells from alveolar tissues were prepared from 1-year-old pigs and the growth kinetics of avian H5 influenza viruses in these cells was investigated. H5N1 virus significantly induced cell death, especially apoptosis, in porcine airway epithelial cells although these three viruses induced similar cell toxicity in chicken embryonic fibroblasts, suggesting that avian H5N1 influenza virus leads severe cell death in mammalian airway epithelial cells due to the induction of apoptosis. A cell clone #21 is a long-term producer of the infectious SARS-CoV, although the incorporation rate of spike (S) protein into virions is significantly lower. Sequencing analysis of the viral structural proteins revealed four and one amino acid substitutions in the S and membrane (M) proteins, respectively. We demonstrated, using a viral-like particle formation system, that the S mutations were involved in lower incorporation of the S protein into virions, although the M mutation disrupting the glycosylation was not involved in this phenotype. Further mutational experiments identified two substitutions, Y442C and L472F, within the receptor binding domain that could be critical for the reduced S incorporation, as well as reduced binding affinity between the S and ACE2 receptor. Thus, these two amino acid substitutions might lead to a conformational change in the S protein, resulting in reduced incorporation into viral particles.
E型肝炎ウイルスの疫学的研究と動物感染モデルにおける解析, 2005, ウイルスの感染動態と環境汚染モニタリングの検討

Five 1-day-old neonatal mice were exposed by oral inoculation to the homogenized brain of a scrapie-affected sheep. In the small intestine of all mice 1 hour after inoculation, immunoreactivity with antinormal prion protein (PrPc) antibody was seen in the cytoplasm of viilus enterocytes. This finding suggests transmission of abnormal PrPsc into the cytoplasm of enterocytes. In control mice treated with normal sheep brain, no PrPc signal was seen in enterocytes of the small intestine. Immunopositivity for neurofilament protein and glial fibrillary acidic protein was seen in the cytoplasm of enterocytes of mice inoculated with scrapie and normal sheep brain. This suggests that the enterocytes of neonatal mice can absorb PrPsc and other macromolecular proteins of the sheep brain affected by scrapie and may be more important than previously thought as a pathway for PrPsc transmission in neonatal animals. After 7 days to 3months of oral inoculation to the homogenized brain of a scrapie-affected sheep, immunoreactivity with antinormal prion protein (PrPc) antibody was not seen in the cytoplasm of villus enterocytes and other tissues in the mucosa. The results suggest that the term of the transmission of abnormal priors protein (PrPsc) ineeds over 3 months and another system of the transmission is present in the mucosa of mouse. Lam inine reseptor (LR) and normal prion were more frequently present in the cytoplasm of enterocytes of suckling mice and young cattle than adult mice and cattle, thus young animals have the higher risk of transmission of abnormal prion than adult animals, via the gastrointestinal menbrane.

Overexpression of PML has been shown to affect the replication of several RNA viruses, including influenza and the complex retrovirus human foamy virus (HFV). Various viruses induce disorganization of the PML NBs, which is thought to be part of a general viral strategy to counteract IFN action. The mononegavirus Boron disease virus (BDV) can persistently infect neurons and astrocytes in the rat central nervous system (CNS) in the presence of a robust IFN response. Replication and transcription of BDV occur in the nucleus, a unique feature among known animal mononegaviruses. BDV antigens colocalize with PML, and persistence of BDV induces PML NBs disorganization suggesting a possible mechanism whereby BDV counteract the IFN response. Vero cells do not produce IFN, hence facilitating the evaluation of PML antiviral activity in the absence of other IFNinduced genes. Overexpression of PML via a recombinant adenovirus (AdPML) induced high levels of PML NBs structures in Vero cells, but did not exert any antiviiral activity against BDV Virus titers, as well as levels of BDV replication and transcription were similar in Vero cells that were infected with AdPML or mock-infected prior infection with BDV. In contrast, and consistent with a previous report, infection with AdPML resulted in about 1000-fold decrease in the production of infectious VSV. NBs have been implicated in hormone signaling, and glucocorticoids are potent modulators of astrocyte function. The resistance of BDV to the antiviral action of PML together with BDV mediated disorganization of PML NBs may contribute to disturbances in CNS function associated with BDV persistence. PML is required for p53 pro-apoptotic signaling, as well as growth arrest inducing pathways. BDV persistence provides increased resistance to program cell death following exposure to Camptothecin. This effect was associated with impaired expression of the pro-apoptotic genes Caspase 3 and Bax. Protection from apoptosis was also observed in cells transfected with BDV N, Together these findings suggest that BDV N might be a repressor of apoptosis, an activity that could contribute to the exquisite ability of BDV to persist in brain cells.

It is pointed out that fatty liver is closely linked to that pathogenesis. Tumor necrosis factor ct (TNFα) secreted from fatty cell is considered to be the most important factor in the insulin resistance. We designed two experiments for induction of fatty liver. In experiment-1, cows were fasted for 4 day to be fallen into negative energy balance. Moreover, as experiment-2, cows were administrated ethionine, analogous substance of methionine, to reduce the production of proteins in the liver. Insulin resistance and TNF α of used cows were analyzed in each experiment. Experiment-1 1. Liver triglyceride (TG) content was apparently increased after fasting. 2. The increase of insulin resistance was observed in the development of fatty liver. 3. Serum concentration of TNFα was not significantly changed in the fasted cow. 4. In micromorphological analysis of liver, hepatic basal lamina in the cow with fatty liver remarkably became thicker compared to that in control. Experiment-2 1. Liver TG contents were remarkably increased in ethionine-treated cows. 2. Serum concentration of apolipoprotein B-100 was significantly decreased in the development of fatty liver. 3. Hepatic lipidosis apparently induced insulin resistance. 4. Hyperglycemia and hyperinsulinism were observed in treated cows. 5. The apparent association of fatty liver with serum concentration of TNFα was not detected. From these results, two experiments were suggested to be effective for induction of fatty liver. Although TNFα of sampled liver tissues were analyzed biochemically and histologically, its relevance to hepatic lipidosis was not detected. We are planning to improve TNFα determination and histological specimens to scrutinize these experiments.

The serological study disclosed the presence of antibodies only to BDV- p40 or -p24 in 19.2 % (24/125) and 3.2 % (4/125) of culled horses, respectively. Antibodies to both BDV -p40 and -p24 were found in 24.0 % (30/125) of these horses. Of particular note was the finding that locomotorium disorders were detectable at a significantly higher rate in BDV antibody positive horses than that in the seronegative horses. These results imply that BDV infection may possibly contribute to an increase in the incidence rate of locomotorium disorders in racehorses. 2.Three central nervous systems collected from three horses with clinical wobbler disease and the Borna disease virus (BDV) positive antibody were investigated histopathologically. Immunohistochemical and in situ PCR examinations were performed for BDV antigens and BDV-RNAs. In their cerebellums, severe glial fibrosis was observed in the medulla and partly cortex. However, inflammatory changes such as perivascular cell infiltration with lymphocytes and macrophages were not observed in any areas. BDV antigens (p40 and p24) in the neuron and neuropil were not observed with immunohistochemical examinations. In situ PCR methods were revealed the minimal presence of BDV-RNAs in the cytoplasm and nuclei of a few number of neuron.

This innovative animal model system greatly facilitated in vivo research for erythroparasitic protozoa that are in nature not infectious to mice. However, the substitution of C.B-17 scid mice with RBCs of some animal species, such as humans and horses, is proved to be very difficult, because of rapid clearance of RBCs transfused in C.B-17 scid mice. In recent years, attempts have been made to develop better SCID mice by changing the genetic background from C.B-17 into the other mice strains, and the NOD-scid mice (the non-obese diabetic mice having the scid mutation) were reported to have significantly improved hematopoietic xeno-transplant acceptability. In the present study, therefore, we examined acceptability of xenogeneic RBCs in NOD/shi-scid in comparison to C.B-17 scid. Clearance tests with fluorescent labeled RBCs of human, dog, horse and cattle showed that life spans of all these RBCs were significantly longer in NOD-scid than in C.B-17 scid mice. In addition, the ability of xeno-RBCs clearance in C.B-17 scid mice was greatly enhanced by some factors, such as optimistic infections, macrophage activating stimuli and aging, whereas such enhancement was poorly observed in NOD-scid mice. These lines of evidence collectively suggest that NOD-scid mice serve as more suitable SCID mice for preparation of the RBC-substitution model than C.B-17 scid. However, NOD-scid mice were less capable of eliminating mouse RBCs upon administration of anti-mouse RBC antibodies, which was effectively used in C.B-17 scid mice to assist rapid RBC-substitution. We were able to demonstrated that NOD-scid mice can easily substituted with human RBCs, and that the human RBC-substituted SCID mice served as an exellent tool for isolation of parasites from a human babesiosis case, which was found for the first time in Japan.
Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research Grant-in-Aid for Encouragement of Young Scientists (A), ボルナ病ウイルス感染動物における発病機序解明の基礎的研究, 1998, 1999, 萩原 克郎, 本年度は昨年度に引き続き、ボルナ病ウイルス(BDV)の疫学調査、特に様々な疾病に罹患し病理解剖に付された馬138頭について抗体陽性率と疾病分類を調査した。まず始めに、ウエスタンブロット法による血清中BDV-p40とp24に対する抗体検査を行なった。その結果、病理解剖馬の陽性率は、BDV-p40 28.3%(39/138)、BDV-p24 14.4%(20/138)、両抗原に対して10.9%(15/138)が陽性であった。その疾病別内訳は、運動器疾患馬でBDV-p40 43.6%(17/39)、BDV-p24 45.0%(9/20)、神経系疾患でBDV-p40 12.8%(5/39)、BDV-p24 15.0%(3/20)、消化器系疾患で、BDV-p40 23.1%(9/39)、BVD-p24 15.0%(3/20)、その他の疾患では、BDV-p40 20.5%(8/39)、BDV-p24 25.0%(5/20)を示し、p-40,24いずれの抗原に対しても運動器疾患馬で40%以上の陽性率が認められた。さらに、顕著な歩行異常を呈した一例の脳内BDV-RNA(p24)検出をRT nested-PCR法で行った結果、脳内の数箇所からBDV-p24が増幅され、ISH及びIHCによる組織内のBDV-RNA、抗原検索により脳神経細胞にBDV陽性シグナルが同一部位で認められた。そこで、脳内抗原陽性部位におけるIL-4、IL-10、IL-12、TNF-α及びINF-γのmRNA遺伝子発現をRT-PCR法で調べた結果、非感染個体の同一部位と比較して抗原陽性部位におけるTNF-αとIFN-γ mRNA遺伝子発現が亢進していた。一方、IL-12 mRNA遺伝子の発現は、BDV陰性個体で発現レベルが増加していた。また、IL-4とIL-10mRNA発現は、BDV感染の有無にかかわらず認められなかった。一般的に、TNF-αは、アストロサイトやミクログリアから産生されオリゴデンドロサイトに傷害性に働き、炎症性脱髄のエフェクターと考えられ、アストロサイトにMHC class I抗原を誘導する。IFN-γはオリゴデンドロサイトにMHC class I抗原を誘導することが知られ、その結果細胞傷害性T細胞の標的になる。本症例は、BDVの感染と脱髄、神経食現象が観察されていることから、上記のサイトカインが関与していることが示唆され、BDVの発症にこれらサイトカインの産生が引き金になっている可能性があり、BDV感染とサイトカイン産生の関係について詳細な研究が必要と思われる。
