Antimicrobial Susceptibility of Escherichia coli and Klebsiella spp. Isolated from Clinical Bovine Mastitis in the Ishikari Area of Hokkaido, SAKEMI Yoko; OKAKOI Masaaki; SHINODA Kojiro; MURAMATSU Yasukazu; UENO Hiroshi; TAMURA Yutaka, Journal of the Japan Veterinary Medical Association, 63, (3) 215 - 218,   2010 03 20 , 乳房炎由来細菌の薬剤感受性状況と、薬剤耐性化による公衆衛生上のリスクを評価する目的で、平成18年6月〜9月に北海道石狩地域で発症した牛乳房炎由来Escherichia coliおよび、Klebsiella属菌について薬剤感受性試験を行った。抗菌剤は、動物用および医療用の18種類を供試した。供試した106株のE. coli中29.2%、34株のKlebsiella属菌中91.2%に耐性株を認めたが、E. coliは家畜由来指標細菌の耐性率より低く、E. coli、Klebsiella属菌ともに二次選択薬である動物用抗菌剤に対してすべて感受性であった。したがって、該当地域における牛乳房炎由来E. coliおよびKlebsiella 属菌の人の健康への直接的な影響は少ないと考えられた。

To verify the possibility, this study was conducted mainly on parasites that included protozoan, helminth and arthropod, and other pathogens including Rickettsia and the Borna virus of 50 free ranging Japanese macaques in the peninsula. Consequently, 5 arthropod and nematode parasite species common to the Japanese macaque, namely Pedicinus obtusus, P.eurygaster, Streptopharagus pigmentatus, Strongyloides fulleborni, and Trichuris trichiura, were obtained in this survey. It is the first record of sites where Pedicinus spp. are seen in a host species.Coccidia and Babesia sp. were not obtained. Coxciella and the Borna virus from the same host samples will be published elsewhere.

Seroepidemiological Study of Coxiella burnetii in Cervus nippon in Northern Japan, YASUMOTO Morihiro; YANASE Tsuyoshi; MURAMATSU Yasukazu; UENO Hiroshi; MORITA Chiharu, Japanese Journal of Zoo and Wildlife Medicine, 2, (2) 101 - 106,   1997 09 01 , Q-fever, caused by Coxiella(C) burnetii, is one of the most important zoonoses. In the present study, the antibody against C.burnetii was analyzed by indirect immunofluorescence antibody(IFA) test in 168 individuals of Japanese deer (Cervus nippon) to determine the prevalence of Q-fever in wild deer of northern Japan. Specific antibody against C.burnetii was found in 72(42.9%) deer, including 24 of 46(52.2%) in Hokkaido and 48 of 122(39.3%) in Tohoku. In Toya and Shari, the positive rate between adult(66.7%) and immature animals(17.6%) was significant (p<0.01). In adult deer, a significant difference was also noted between Hokkaido and Tohoku(p<0.01).

Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, The research for ansector of methicillin-resistant Staphylococcus pseudintermedius among dogs in Japan, 20140401, 20180331, Ishihara Kanako; TAMURA Yutaka; MURAMATSU Yasukazu, The recent appearance of methicillin-resistant Staphylococcus pseudintermedius (MRSP) is a concern for both veterinary and human healthcare. The current study compared the molecular characteristics of 43 MRSP isolates from dogs in Japan with those of MRSP from previous reports using multilocus sequence typing, SCCmec typing, and detection of antimicrobial resistance genes. Three related clonal lineages, ST71, ST169, and the newly registered ST354, were observed amongst SCCmec II-III isolates from Japan, despite MRSP SCCmec II-III isolates being thought to belong to a single clonal lineage. The majority of SCCmec II-III isolates belonging to ST169 (9/11) and ST354 (3/3), but not ST71 (0/11), harboured tetM. Since the allele sequences of two loci (ack and sar) differ between ST71 and ST169 isolates, ST71 and ST169 clones have been derived from a common ancestral clone. ST354 is likely a variant clone of ST169.

Rabies virus (1) Indonesia : The N gene was detected from a dog brain that suffered rabies, and resembled that of isolates in China and Philippines. (2) Vietnam : The N gene was not detected from bat brain, but neutralizing antibody for rabies virus was found in dog serum. 2. Q fever rickettsia (1) Thailand : An antibody for Q fever was detected from chicken and cattle serum, and detected Coxiella DNA from ticks attached to cattle. (2) Indonesia : An antibody for Q fever was detected from chicken and cattle serum. (3) Vietnam : An antibody for Q fever was detected from chicken and cattle serum, and Coxiella DNA was detected from chicken and cattle. 3. Drug resistant bacteria (1) Thailand : A lot of multiple drug-resistant Enterococci were isolated from chicken feces. (2) Indonesia: Multiple drug-resistant Enterococci was isolated from chicken feces, and a lot of vancomycin resistant Enterococci (VRE) were isolated. (3) Vietnam : A lot of multiple drug-resistant Enterococci were isolated from chicken and cattle feces. From these results, rabies virus, Coxiella burnetti and drug resistant bacteria including VRE spread in the southeast asian countries which surrounded our country, and the possibility exists to invade our country through an animal and food.
Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Experimental transmission of abnormal prion protein (PrPsc) in the small intestinal epithelial cells of neonatal mice, 2003, 2004, TANIYAMA Hiroyuki; HAGIWARA Katsurou; MURAMATSU Yasukazu, Using an immunohistochemical method, we attempted to detect the transmission of abnormal prion protein (PrPsc) to the enterocytes of the small intestine of neonatal mice by oral exposure with sheep brain affected by scrapie. Five 1-day-old neonatal mice were exposed by oral inoculation to the homogenized brain of a scrapie-affected sheep. In the small intestine of all mice 1 hour after inoculation, immunoreactivity with antinormal prion protein (PrPc) antibody was seen in the cytoplasm of viilus enterocytes. This finding suggests transmission of abnormal PrPsc into the cytoplasm of enterocytes. In control mice treated with normal sheep brain, no PrPc signal was seen in enterocytes of the small intestine. Immunopositivity for neurofilament protein and glial fibrillary acidic protein was seen in the cytoplasm of enterocytes of mice inoculated with scrapie and normal sheep brain. This suggests that the enterocytes of neonatal mice can absorb PrPsc and other macromolecular proteins of the sheep brain affected by scrapie and may be more important than previously thought as a pathway for PrPsc transmission in neonatal animals. After 7 days to 3months of oral inoculation to the homogenized brain of a scrapie-affected sheep, immunoreactivity with antinormal prion protein (PrPc) antibody was not seen in the cytoplasm of villus enterocytes and other tissues in the mucosa. The results suggest that the term of the transmission of abnormal priors protein (PrPsc) ineeds over 3 months and another system of the transmission is present in the mucosa of mouse. Lam inine reseptor (LR) and normal prion were more frequently present in the cytoplasm of enterocytes of suckling mice and young cattle than adult mice and cattle, thus young animals have the higher risk of transmission of abnormal prion than adult animals, via the gastrointestinal menbrane.
Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Molecular phylogenic study of Ehrlichia spp. microorganisms as the etiological agents of emerging diseases in Japan, 2002, 2004, MURAMATSU Yasukazu, 1)Ehrlichial gene was detected from small rodents trapped in a Lyme disease-endemic area in Hokkaido, the northernmost island of Japan. Primer pairs of 168 rDNA targeting the genus of Ehrlichia and other region of the 16S rDNA specific for E. chaffeensis and E. muris were used for identification. The DNA fragment specific for 16S rDNA of E. spp. was detected from 4 of 94 Apodemus. speciosus mice (positive rate 4.3%) and 5 of 73 Clethrionomys rufocanus bedfordiae mice (positive rate 6.8%). The nucleotide sequence of the amplified 168 rDNA fragment was most similar to those of E muris-like ehrlichia, E spp HF565 and Shizuoka-36 originating in the northeastern part of Japan. In phylogenic analysis based on 16S rDNA sequences, these northeastern and southwestern groups of E. murrslike ehrlichia made a cluster with E. muris group microorganisms. These results suggest that there are two groups of E. murislike microorganisms in Japan. 2)A novel genotype of Ehrlichia rummantium, the causative agent of heartwater, was genetically detected in the Sudan. The new genotype was provisionally named as "Gedaref". The results of phylogenic analyses on the regions of pCS20 and major antigen protein I (map)) genes revealed that the ehrlichia) strain belongs to E. ruminantium-group, but does not correspond with any known strains of E. ruminantium. Furthermore, as the results of phylogenic analyses on map), "Gedaref" made clusters with several strains of E. ruminantium other than Um Banein, which had been known as the sole strain of E. ruminantium in the Sudan. This work provided the evidence that not only Um Banein strain but also the other strain exist in the Sudan.
Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Etiological and molecular genetic surveillance of scrapie in Mongolia, 2000, 2002, HORIUCHI Motohiro; MURAMATSU Yasukazu; ISHIGURO Naotaka, Mongolia has a far long history for sheep-farming and is now farming over twelve million sheep. In the future, Mongolia has a potential for the major provider of livestock products among the world. Thus surveillance of prion diseases in animal in Mongolia will provide important information to evaluate the safety of livestock products in the country. During the research term, we visit to central, west, and east part of Mongolia, we did not get a positive indication on the existence of scrapie-suspected sheep in hearing. We also checked PrPSc in the medulla oblongata from sheep showed neurological symptoms, but all were negative for PrPSc. In order to investigate the diversity of PrP gene of sheep in Mongolia, we collected a total of 470 venous blood samples from 9 different breeds (Khalkh, Yeroo, Orkhon, Khangai, Sartuul, Govi-altai, Bayad, Darhad, and Symber-Karakul) and purified genomic DNA. PrP genotype (amino acid type) was determined by direct sequencing of PCR-amplified fragment corresponding to PrP open reading frame. With regard to PrP genotype at codon 171 determining susceptibility to scrapie, sheep carrying 171-Gln/Gln genotype that is associated with the susceptibility to scrape, varied from 58 to 97% among the breeds. Sheep carrying 171-Arg/Arg or 171-Arg/Gln genotype that confers resistance to scrapie infection were less abundant, especially only a small percentage of sheep possessed homozygous for 171-Arg alle. These results indicate sheep in Mongolia is composed rather of scrapie-susceptible population, so continuous surveillance as well as preventive measures against prion-infected animal are important to evaluate the risk of prion disease. These results are also useful for performing a selective breeding program that produce scrapie-resistant flock.
Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Investigation of Listeria monocytogenes in organic farming environments based on detection of VBNC cells of the organism., 1998, 2000, UBNO Hiroshi, Listeria monocytogenes has a possibility to form VBNC (Viable But Non-Culturable) cells in natural environments, although knowledge of this phenomenon in the bacteria is very limitted. The author found that the suitable conditions for detecting VBNC cells of a test strain of L.monocytogenes were incubation at a concentration of 1/2 to 2/3 MIC in brain heart infusion for 4 to 8 hours after contact with levofloxacin. Because these conditions did not allow division of the test organism, but allowed formation of elongated VBNC cells, stained by Giemsa solution, in a liquid medium. About 10^7/ml of viable test organisms were added to sterilized soil solution, PBS and distilled water, and settled for 56 days at 0℃. During this period, plate counts and total bacterial counts were almost constant in sterilized soil solution, whereas VBNC cells were frequently (43% of numbers of total cells) found in 7 days after settlement. The similar results were obtained in PBS, although VBNC cells were seldom observed in distilled water. These results suggest that entry of L.monocytogenes organisms into VBNC state occurs at cold and starved conditions. After inoculating 6×10^7/ml of viable organisms in sterilized soil solution, the solution was concentrated by ten using PBS.Plate counts increased to 4.7×10^8/ml in the solution and almost same numbers (2.1×10^8/ml) of total bacterial counts were observed by an enzyme-linked immuno-sorbent assay (EIA) using peroxydase labeled L.monocytogenes antibodies. On the other hand, there was no stained organism in a sample taken from un-sterilized soil solution that yielded a plate count of 9.2×10^5/ml. These results suggest that L.monocytogenes in soil could be detected specifically by using the EIA.
日本学術振興会, 科学研究費助成事業, リポ多糖解析に基づくcoxiella burnetiiの分子疫学的研究, 1997, 1998, 村松 康和, 北海道のウシの生乳から、マウスを用いて継代・分離した2株のCoxiella burnetii、Nine Mile II相菌(American Type Culture Collectionより購入;ATCC VR-616)、Califonia株(同;ATCC VR-614)、およびPricilla株(岐阜大学;平井克哉教授より分与)を発育鶏卵卵黄嚢内接種により大量培養した。次いでWilliams,J.C.et al.(1981)の方法に従いRenografinの密度勾配を用いた遠心分離によりC.burnetiiを精製した。 得られたコクシエラの精製菌を用いてSDS-PAGEを行った。次いで銀染色、およびWesternblot analysis(WBA)により、本研究による分離菌と、既存の各菌株との蛋白、および抗原性状に関する比較を行った。 銀染色の結果、本研究で得られた分離株には、比較に用いたNine Mile H相菌株、およびCalifornia株と異なる、株特異な蛋白画分は認められなかった。また、用いた全てのコクシエラで全蛋白の泳動像に差異は認められなかった。これらの結果は、国内外における以前の報告を裏付けるものである。 抗コクシエラ・マウス血清を用いたWBAの結果、分離2株中、1株において、他のコクシエラとは異なる抗原性状が認められた。現在、糖鎖の性状について検討中である。 血液吸着濾紙により採取した見かけ上健康なネコの乾燥濾紙血液を用いて、間接蛍光抗体法、およびPCRを行った。その結果、今回初めてPCRにより乾燥濾紙血液からのコクシエラ、およびネコひっかき病の主要な原因リケッチアであるBartonella henselaeに特異なDNAの検出に成功した。この事は乾燥濾紙血液サンプルが広範囲にわたるコクシエラの疫学調査に有用であることを示している(現在投稿準備中)。
日本学術振興会, 科学研究費助成事業, PCR-ELISAによるQ熱の診断に関する研究, 1995, 1995, 村松 康和, BGM細胞に0.001〜0.1%濃度の界面活性剤Triton X-100 (TX100)を加え、鏡検により細胞の破砕を観察した。Coxiella burnetii Nine Mile II相菌(ATCC VR-616)の培養上清(細胞外の菌体が主体)、及び感染BGM細胞(細胞内の菌体が主体)を別々に回収した。 培養上清について各々0.001〜0.01%濃度のTX100を加えた試料と無添加の試料を遠心後、PCRを行った結果、無添加の場合と検出感度に差は無かった。 感染細胞では0.01、0.02%のTX100を加え遠心した試料、免疫鉄ビーズで菌体を回収した(この方法についてはVeterinary microbiologyに掲載決定)。試料、及びTX100添加後、免疫鉄ビーズで菌体を回収した試料でPCRを行った。0.01%濃度のTX100を加えた試料では、TX100無添加で免疫鉄ビーズにより菌体回収した試料と比べPCRの結果、検出感度の上昇が見られた。また、0.01%TX100は免疫鉄ビーズと菌体の反応を阻害しないことが判った。牛乳の場合、0.01%TX100のみ添加では遠心後に大量の堅固なpelletが残り、PCRサンプル調整の妨げになると共に検出感度が低下した。牛乳からのPCR-ELISAサンプル調整では0.01%TX100添加+免疫鉄ビーズによる菌体回収で検出感度が良かった。 ウシ60頭の生乳から、この方法によりサンプル調整し、PCR-ELISAによりC. burnetii検出を行った。PCR-ELISAは先に提出した申請書に記載した方法に準じて行った。10頭が陽性で(陽性率16.7%)、うち5頭が強陽性であった。PCR-ELISAは高感度に、多数の試料の判定が出来る利点を持つ。(以上の結果について第121回日本獣医学会で発表予定。Applied and environmental microbiologyに投稿準備中。)
