Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Establishment of the cultivated corneal tissue for the corneal transplantation, 20140829, 20160331, Nam Eunryel, Collagen alignments induce cell alignments that can improve biological functionalities such as cornea transparency. Since in-vitro alignment of collagen nanofibers has been difficult, mechanical stresses have been applied to align collagen and cells together during cultivation. However, the stresses during cultivation often induced decreased tissue stability. In this work, we show that 1) the collagen nanofiber alignment is optimized by changing of cyclic stretch conditions such as frequency, strain, and time, and 2) the fabricated collagen substrate aligns cells without any mechanical stresses during cultivation, thus even enabling corneal stromal cell alignments which are known to be difficult at in-vitro. Moreover, collagen nanofibers are self-aligned on 3D surfaces by applying cyclic stretch during the gelation process of collagen solution. We believe that this method for collagen and cell alignments contributes to provides microenvironmentson 3-dimensional surfaces of organs.
