Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Preventive effects of marine carotenoids on chronic inflammation-related diseases through regulation of cell and tissue communication, 20110401, 20140331, HOSOKAWA Masashi; YASUI Yumiko, Fucoxanthin (Fx) suppressed chronic inflammation in white adipose tissue by the inhibition of macrophage and T cell infiltration. Further, Fx down-regulated mRNA expression of chemokines and adipokines, which recruit immune cells and induce insulin resistance. In diabetic/obese mice fed Fx, GLUT4 activation was observed in skeletal muscle through the improvement of hyperglycemia. On the other hand, Fx and astaxanthin (Asx) extracted from H. pluvialis prevent DSS-induced colitis and colon carcinogenesis in mice. In colon of the mice fed Asx, mRNA expression levels of inflammatory cytokines tended to decrease compared to control mice. In addition, Asx down-regulated mRNA expression of inflammatory cytokines in macrophage-like cells and colon cells stimulated by the culture medium of activated macrophages. These results suggest that marine carotenoids prevent chronic inflammation and its-related diseases by the regulation of cell and tissue communication via chemokines and cytokines.
Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Analysis of the relation between prostate carcinognenesis targeted stem cells and inflammation, 2009, 2011, SUGIE Shigeyuki; TANAKA Takuji; YASUI Yumiko; OYAMA Takeru, In this study, the relation between prostate carcinogenesis and stem cell or inflammation is analyse in animal experimental carcinogenesis model targeted stem cells associated with prostatitis. Any prostate neoplasma were not found in any castrated rats, although prostate neoplasms was observed only in non-castration groups. From these results, the origin of cancer was assumed to be lost by castration. TP had a suppressive effect in PhIP prostate carcinogenesis. he hypothesis which Prostate cancer originated from stem cell. The lobes with inflammation were different the lobes with PIN. The modifying effect of inflammation for prostate carcinogenesis is not clear, yet. Further consideration will be necessary.
Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Involvement of obesity in oral carcinogenesis using animal models, 2008, 2010, TANAKA Takuji; SUGIE Shigeyuki; YASUI Yumiko, We investigated whether obesity influences oral (tongue) carcinogenesis using obese model animals, such as db/db and ob/ob mice. The results indicated that the obese mice were highly susceptible to 4-NQO-induced tongue and esophageal carcinogenesis when compared to their wild types (WT) of mice. In addition, the susceptibility was high in db/db mice in comparison with ob/ob mice. The immunohistochemical expression of pro-inflammatory cytokines (IL-1α, IL-1β, interleukin-4, interleukin-6, interleukin-8, and TNF-α) in the tongue squamous cell carcinomas which developed in the db/db mice were significantly greater than that developed in the WT mice (p<0.05-p<0.01). However, there were no significant differences in the expressions of NF-κB, Stat3, and Nrf2 between the obese and WT mice. Our results may suggest that overexpression of these pro-inflammatory cytokines plays an important role in highly susceptible to 4-NQO-induced tongue and esophageal carcinogenesis in obese mice.
Japan Society for the Promotion of Science, Grants-in-Aid for Scientific Research, Preventive effects of marine carotenoids on life-style-related diseases based on anti-inflammatory effects as molecular targets, 2008, 2010, HOSOKAWA Masashi; YASUI Yumiko, Astaxanthin exhibited inhibitory effects on dextran sodium sulfate-induced colitis and inflammatory-related colon carcinogenesis in mice. In colon of the mice fed astaxanthin, mRNA expression levels of inflammatory cytokines such as TNF-α and IL-1β were suppressed through inhibition of NF-κB activation. After feeding for 20 weeks, colonic mucosal ulcers, dysplastic crypts and colonic adenocarcinoma were inhibited by 200 ppm astaxanthin-containing diet. In addition, astaxanthin, pectenolone, alloxanthin and diatoxanthin isolated from marine organisms attenuated excessive mRNA expression of inflammatory cytokines and COX-2 and iNOS in macrophages activated by LPS. On the other hand, dietary 0.2% fucoxanthin markedly suppressed mRNA expression of inflammatory cytokines such as TNF-α and IL-6 to induce insulin resistance and inhibited macrophage infiltration in white adipose tissue through suppression of MCP-1 mRNA expression after 4 weeks feeding. Further, fucoxanthinol, which is one of fucoxanthin metabolites, also regulated cross talk of adipocytes and macrophages via palmitic acid and TNF-α. These results suggest that anti-inflammatory effects are molecular mechanism for anti-obesity and anti-diabetic effects of fucoxanthin.
日本学術振興会, 科学研究費助成事業, 炎症性大腸発がんモデルマウスを用いた共役リノレン酸の発がん阻止効果に関する研究, 2006, 2007, 安井 由美子, 平成19年度は平成18年度に続き、20週目の剖検時における病理組織学的解析と、採取した大腸粘膜を用いて、発がんに関与する遺伝子の発現をリアルタイムPCRにて解析を行った。剖検時、いずれの実験群にも体重および肝重量に有意な差は見られなかった。一方、大腸の長さに関してはAOM+1.5% DSS+0.1% PGO群において、その他の群よりも、大腸が有意に短かった。大腸粘膜潰瘍と異型陰窩の発生に関して、AOM+1.5% DSS群と比較して、PGO処理によりいずれの濃度においても有意な(P<0.001)抑制が観察された。大腸腺腫の発生に関してはAOM+1.5% DSS群(0.9±1.10)で50%の発生が見られ、1% PGO処理(0.3±0.71)により抑制傾向が見られたものの有意な差は得られなかった。一方、腺がんの発生に関しては、AOM+1.5% DSS群(2.9±1.29)で90%の発生が見られ、いずれの濃度においても有意抑制効果が見られた(0.01% PGO:0.9±0.78,P<0.01,0.1% PGO:1.2±1.14,P<0.05,1%PGO:0.9±1.27,P<0.01)。さらに、大腸粘膜における発がん関連遺伝子(PPARγ、COX-2)の発現をリアルタイムPCRで解析した。その結果、20週時においてPGO処理によるPPARγの発現増加、COX-2の発現抑制の傾向が得られた。以上の結果から、CLNを高濃度含有するPGOは、炎症を背景とする大腸がんの発生を抑制することが明らかとなった。また、腺腫の発生に有意な差は見られず、腺がんの発生において顕著な抑制が見られたことから、特に腺腫から腺がんに進行するプログレッションの段階にPGOが作用することが示唆された。
