HPCD子豚へのSalmonella enterica serovar Choleraesuis実験感染, 溝呂木 聖子; 矢ヶ部 陽子; 芝原 友幸; 秋庭 正人; 楠本 正博; 佐久間 晶子; 中條 満; 新開 浩樹; 上西 博英; 宗田 吉広, 日本豚病研究会報, (63) 6 - 11,   2014 02 , Toll-like receptor5(TLR5)は細菌のべん毛タンパク質(flagellin)を認識するパターン認識受容体で、豚ゲノムの解析においてサルモネラのflagellin及びSalmonella enterica serovar Choleraesuis(SC)の認識を低下させるTLR5一塩基多型(single nucleotide polymorphism: SNP) (C1205T)が報告されている。当該SNPがヘテロ型のランドレース豚の雌雄を交配して得られる妊娠母豚の子宮切除術によって作出されたhysterectomy produced colostrum deprived(HPCD)子豚へSCを無菌ミルクに混ぜて2回の経口実験感染を行った結果、HPCD子豚へのSC経口感染により、これまで再現が困難であった下痢や敗血症を特徴とする、豚サルモネラ症の病態モデルの再現ができた。しかし当該TLR5 SNPのSC感染に対する影響については、感染初期での排菌量に有意差は認められず、生存日数への影響についても2回の実験からは確かめられなかった。その理由としてHPCD子豚ではflagellin刺激に対するIL-6応答の顕著な低下が原因である可能性が示唆された。

ヒトと動物に共通する腸内細菌のストレス耐性, 中澤 宗生; 秋庭 正人; 佐藤 国雄, 畜産の研究, 62, (2) 252 - 254,   2008 02 , 人獣共通感染性腸内細菌である腸管出血性大腸菌(EHEC)O157を含む志賀毒素産生性大腸菌、サルモネラ、エンテロバクターなどの各種ストレスに対する耐性を調べた。EHEC O157:H7、志賀毒素産生性大腸菌、一般大腸菌はサルモネラやエンテロバクターに比べ高い酸耐性を示した。EHEC O157の酸耐性は一般大腸菌と比較しても有意に高かく、熱耐性でも、他の大腸菌やサルモネラより高い数値を示した。ヒトのEHEC O157:H7の感染菌量は100個未満であり、少量菌で感染が成立する要因の一つとして本菌のもつ強い酸耐性が考えられた。また、サルモネラの熱耐性では、感染宿主域が狭く種特異性が高い血清型Dublin、TyphisuisおよびPullormは熱ストレスに対してまったく抵抗できなかったが、宿主域が広く種特異性の低い血清型Enteritidis、Typhimurium、Agona、Infantis、SenftenbergおよびVirchowは0.001%〜30.0%の生残率を示した。サルモネラ食中毒の主要血清型であるこれら菌型のもつストレス耐性能は、環境や食品中での生存性に関与する要因の一つである考えられた。以上のように、腸内細菌のストレス耐性は菌種よる違いがあるが、強いストレス耐性能を有する菌型が存在することから、これらの特性を考慮した制御法が必要である。
